Abstract


Objective: To investigate the Hsp70- peptide complexes isolated from gastric carcinoma and pulsed dendritic cells ( DC ) , was observed in the in vitro anti-tumor effect , DC -mediated tumor immunotherapy provide a theoretical experimental basis . Methods: GM - CSF , IL - 4 stimulation , cultured gastric cancer patients peripheral blood mononuclear cells ( PBMC ) proliferation to produce a large number of DC , HSP70 - peptide complexes extracted from clinical gastric cancer tissues using Sephadex G - 100 gel chromatography , SDS- polyacrylamide gel electrophoresis and Western - bolt identification of its molecular weight was measured in parallel . With complex modified DC, DC supernatant IL-12, TNF-α, IL-1β content detected by ELISA modified DC and not Hsp70- peptide complexes modified with DC activated autologous mixed T lymphocytes observed in vitro killing effect on gastric cancer cells , not Hsp70- peptide complexes modified DC as control . Results: Sephadex G - 100 gel chromatography of proteins extracted from the clinical gastric cancer tissues by SDS-polyacrylamide gel electrophoresis and Western - bolt identification for HSP70 , PBMC obtained from patients with gastric cancer by GM - CSF , IL -4 cultured and stimulated 7d electron microscopy cells irregular in shape , cell surface asperities , kidney - shaped nucleus and horseshoe-shaped , can be identified as DC . Hsp70 modified DC cytokine secretion of IL-12, TNF-α, IL-1β content (305.02 ± 20.70) pg / ml, (480.61 ± 32.52) pg / ml, (519.60 ± 39.12) pg / ml, while is not Hsp70 modified DC secretion of cytokines IL - 12 , TNF-alpha , IL -1beta content ( 3.81 ± 0.84 ) pg / ml ( 3.50 ± 0.21 ) pg / ml , ( 50.11 ± 2.86 ) pg / ml . between the two significant differences ( P < 0.01 ) . Hsp70 modified DC - activated T lymphocytes in gastric cancer cell lethality ( 93.5 ± 8.2 ) % of mass destruction with the control group ( 6.2 ± 0.3 % ) compared with the significant difference ( P < 0.01 ) . Conclusion: The protein extracted from gastric tissue for HSP70 , the protein retains the original biological activity of Hsp70 protein , tumor antigen . Hsp70 modified DC to produce IL-12, TNF-α, IL-1β cytokine levels were significantly increased , modified DC activation of T lymphocytes has a strong killing effect on tumor cells .
