Abstract


This paper studies a winery in Ningxia mansions scraps - red grape seed extract OPC's as raw material process . The OPC's extracted at a wavelength of 500nm with a spectrophotometer , using vanillin - Determination of acid content . Through a large number of experiments to determine extraction process route has the following steps : 1. Extraction - first defatted with petroleum ether to 60% ethanol as extraction solvent the best, at 50 ℃ under the conditions of solid-liquid ratio 1:7 extraction 30min, extracted for 3 times to obtain a crude product. After ethanol as a solvent central composite experimental optimization, using regression analysis experiment to get the best response to all things technological conditions were : extraction temperature 51.8 ℃, extraction time 30min, solid-liquid ratio of 1:7 , extraction for 3 times concentration of 60.7% , 91.38% extraction rate . 2 , the extraction - precipitation of the crude product after removal of proteins , using ethyl acetate as extractant purification of the crude extract , the optimum conditions were: temperature 0 ℃, ethyl acetate - water volume ratio of 3:1 , extraction for 3 times ; petroleum ether as a precipitation solvent selected , a saturated solution of ethyl acetate at room temperature , with 3 volumes of petroleum ether product is purified , precipitated product was purified twice . 3 , column chromatography - by column chromatography macroporous resin to further optimize the product , determine macroporous resin AB-8 is the best adsorption resin , with 60 % ethanol as eluent , prepared after optimization of process conditions product purity was 92.7% , the yield was 1.87% ( calculated by 100g grape seed ) . 4 , analysis - through the product fumble TLC experiments elect eluent toluene: acetone: acetic acid = 3:3:1 optimum separation efficiency and the product by TLC preliminary analysis by high performance liquid chromatography (HPLC) analysis through the column of the product, determine its composition is consistent with the standard sample . 5 , performance measurement - Since OPC's antioxidant properties of the material, so the oxidation products extracted from experiments , as determined by the fermentation of red grape seed extract antioxidant products significantly higher than Vc and Ve, and the OPC's oxidation resistance at 0 ~ 2000ppm , with the added amount of the increase .
