Abstract

Rice is one of the most important food crops around the world, about one-third of the population of rice as a main food. Therefore, to improve rice production and improve rice quality related to human survival, development and health. Selected by pedigree sexual hybridization, mutation breeding methods, many closely related species of good traits grouped together. However, due to the presence of reproductive isolation, which makes the plant genetic improvement has been limited to a large extent. Plant genetic engineering research began in the 1970s in order to expand the plant gene pool provided new possibilities opened up a new way for the genetic improvement of plants. The plants transgenic research purposes molecular biology and genetic engineering techniques to insert a foreign gene integrated into the receptor genes of plants, and it can be genetic and expression in progeny plants, so that the receptor plants new traits . Obtained since 1983 the first transgenic plants (tobacco), began the era of the use of transgenic technology improved species sexual; Horsh et al 1985 first leaf disc transformation method, the Agrobacterium transformation process greatly simplified from a plant gene translational research have developed rapidly. Short awn barley [Hordeum brevisubulatum (Trin.) Link] Poaceae Hordeum a perennial herb, commonly known as wild barley. Widely distributed in the Northeast, North China, Inner Mongolia, Qinghai and Xinjiang, the advantage is cold, salinity, tillering ability and other characteristics. In recent years, we have short awn barley material, has conducted extensive research in physiological, biochemical characteristics and salt-tolerant mutant. The Na ~ vacuoles regionalization plants in particular is the halophytes adapt Na ~ poisoned a very effective means vacuolar Na to / H ~~ antiporter in this process played an important role. This article was cloned from the short awn barley vacuolar Na / H antiporter gene length cDNA sequence, named HbNHX1. Bioinformatic analysis showed that the, HbNHX1 contains 11 transmembrane region. HbNHX1 phylogenetic analysis of the cloned Na ~ / H ~ antiporter system with vacuolar Na ~~ / H ~~ antiporter homology, approximately in the range of 55.3-74.8%, and the quality membrane-type Na to / H ~~ the antiporter The homology lower, in the range of 18.3-21.8%, description HbNHX1 are typical vacuolar Na ~ / H ~ antiporter. RT-PCR analysis showed that the roots and seedlings in transcriptional level HbNHX1 gene expression, but abundant expression of seedlings; without salt stress-induced expression of the gene at the transcriptional level. Order of further identification HbNHX1 gene function, this article from the short awn barley vacuolar Na cloning to / H ~~ the antiporter gene HbNHX1 build plant effective expression vector pBRNHX Agrobacterium-mediated transformation of important food crops of rice. The results obtained 20 transgenic plants, PCR, PCR-Southern and RT-PCR results show that the HbNHX1 gene was integrated into the nuclear genome of the transgenic rice and expression at the transcriptional level. Laid a good foundation for the stress resistance of rice genetic improvement.
