Abstract


Objective: To investigate curcumin on vascular smooth muscle cells (VSMCs) and its impact on the accumulation of cholesterol sterol regulatory element binding protein -1 (SREBP-1) activity relationships . Methods : Cultured rat aortic VSMCs, different concentrations of curcumin (0,6,12.5,25,50 μmol / L) acting on the cells for 12 hours, then take the optimal concentration 12.5μmol / L curcumin at different times ( 0,3 , 6,12,24,48 h) acting on the cells. HPLC assay of intracellular total cholesterol (TC) levels ; Western blotting Caveolin-1 protein expression . Then 50mg/Lox-LDL effect on VSMCs 48 hours, causing bearing fat cell model , different concentrations of curcumin (0,12.5,25,50 μmol / L) acting on the bearing fat cells for 24 hours, and then take the best concentration of 25μmol / L curcumin at different times (0,6,12,24,48 h) acts on cells bearing grease , high performance liquid chromatography (HPLC) detection of intracellular total cholesterol (TC), free cholesterol (FC) and cholesteryl ester (CE) the content , Western blotting Caveolin-1 and SREBP-1 protein expression of an active fragment . Finally, SREBP-1 protein inhibitors ALLN intervention , 25μmol / L curcumin in VSMCs derived lipid load cells for 24 hours, oil red O staining of intracellular lipid droplets situation , HPLC detection of intracellular TC, FC, CE content , Western blotting active fragment of SREBP-1 , Caveolin-1 protein expression . Results : Different concentrations of curcumin (0,6,12.5,25,50 μmol / L) acting on VSMCs 12 hours, intracellular TC and Caveolin-1 expression levels were reduced ; 12.5μmol / L curcumin at different times ( 0,3 , 6,12,24,48 h) acting on VSMCs, TC contents and intracellular expression of Caveolin-1 showed upward trend after the first cut . Different concentrations of curcumin (0,12.5,25,50 μmol / L) acts on fat cells - derived VSMCs 24 hours and 25μmol / L curcumin at different times (0,6,12,24,48 h) acting on VSMCs derived
