Abstract


Gastric cancer is one of the most common malignant neoplasms in China and its death incidence is gradually increasing in recent years. Studies have shown it is the leading cause of cancer death. The clinical outcome of gastric cancer patients is still very poor with a 5-year survival rate of only 20% in all stages and only 40% of the patients respond to surgical intervention. It is kown that gastric cancer is the result of accumulation of multiple gene mutations that mainly occur at three kinds of genes, which are oncogene, tumor suppressor gene and DNA mismatch repair gene. Most of them are somatic cell variations, including mutation, amplification, rearrangement, deletion or changes of methylation status. The mechanism of gastric cancer is not clear and the therapeutic effect is not good. To study the expression and distribution of RegIV and its relationship with PCNA expression, to investigate possible roles of RegIV in the gastriccarcinogenic mechanism and to provide a new method to diagnosis, therapy, and prognosis assessment.Specimens of gastric cancer and corresponding adjacent normal gastric tissue were obtained from 26 cases who had undergone surgical operations of gastric cancer at the first and the second hospitals affiliated to the Third Military Medical University. All samples were confirmed by pathologyhospital diagnosis. Expressions of RegIV mRNA were dectected by semiquatitative reverse transcriptase polymerise chain reaction in 10 specimens of gastric cancer and corresponding adjacent normal gastric tissue, expressions of RegIV protein were dectected by immunohistochemistry in 26 specimens of gastric cancer and 10 specimens of adjacent normal gastric tissue, and further we investigated the relationships between the expression of RegIV protein and the clinicopathologicparameters. Besides, we dectected the PCNA labeling index and investigated the coexpression of RegIV and PCNA by double-labelling immunofluorescence in specimens of gastric cancer, to explore the possible role of RegIV in proliferation of gastric cancer. The data were analysized by software SPSS11.0 using Fisher’s Exact Test, Crosstabs analysis, Mann-Whitney U Test and Wilcoxon Signed Ranks Test.
