Gastric cancer is one of the most common malignancy in the world. Gastrin is a polypeptide hormone, secreted by G cells of the gastrointestinal tract, and gastrin receptor specific binding, and primarily has the nutritional role of stimulated gastric acid secretion and gastrointestinal mucosa. Gastrin as gastrointestinal peptides showed strong proliferative effect on most normal and malignant transformation of gastrointestinal mucosa, and plays an important role in the process of malignant transformation of the gastric mucosa. A large number of studies have shown that gastrin mediated series of intracellular signal transduction through its receptor, to promote cell division and DNA synthesis, and promoting growth of gastric cancer, colorectal cancer and other gastrointestinal tumors. However, the gastrin mechanism to promote the proliferation of gastric cancer cells remains unclear. Abroad in recent years, studies have shown that Reg Ⅰ protein is highly expressed in gastric carcinoma, gastric carcinoma Reg Ⅰ protein-positive rate of 37.7%. In gastric cancer, Reg Ⅰ protein can be used as growth promoting and anti-apoptotic factors, Reg I-positive early gastric cancer showed higher PCNA labeling index Reg I protein can promote cell growth, inhibit H 2 O 2 -induced gastric cancer cell lines AGS apoptosis. Reg gene expression and differentiation of gastric cancer, invasive growth, as well as the patient's prognosis there is a correlation; the Reg gene-positive patients with gastric cancer, tumor differentiation degree is low, and the prognosis is poor; the invasive gastric Reg gene expression was significantly higher than original bit of gastric cancer. Gastrin can stimulate the Reg gene's expression in gastric cancer cells. The hypergastrinemia patient's stomach tissue Reg gene expression, accompanied by proliferation of gastric mucosal cells. Reg protein can stimulate the growth of cultured gastric mucosal cells, and a dose-dependent and time-dependent manner; direct growth-stimulating effect of gastrin on gastric mucosal cells in culture, but by stimulating the expression of Reg protein, thereby stimulating the stomach mucosal cell growth. Reg protein in gastric tissue, Reg Ⅰ expression chromaffin-like (enterochromaffin-lik celle, ECL) cells in the stomach at the bottom of intestinal. Gastrin REG protein activity for ECL cell stimulating factor. Gastrin and gastrin receptor Reg Ⅰ gene co-expression at home and abroad about gastric tissue and gastric histopathological features of the correlation is rarely reported. Domestic Reg gene few reports of its main targets for colorectal cancer, no study of the gastric cancer. In this study, immunohistochemical techniques, observation of 36 cases of gastric carcinoma, adjacent tissues and gastrin and gastrin receptor expression in normal gastric mucosa; using RT-PCR technology to detect 36 cases of gastric carcinoma, adjacent tissues and Reg Ⅰ gene expression in normal gastric mucosa, analysis of gastrin and gastrin receptor Reg Ⅰ gene expression and pathological features of gastric cancer correlation. To further verify the relationship of gastrin Reg Ⅰ gene expression, the study compared the gastric cancer cell line BGC-823 Reg Ⅰ gene expression changes in the expression of gastrin suppression. The research results will clarify gastrin promoting molecular mechanism of gastric cancer, and to provide new experimental evidence. Materials and methods. The material in this experiment to collect the First Affiliated Hospital of Zhengzhou University, surgical resection and pathologically confirmed 36 cases of gastric carcinoma, 15 cases of cancer adjacent tissues and 11 cases of normal gastric mucosa. Immunohistochemistry gastrin in normal gastric mucosa paracancerous organizations and gastric carcinoma, the positive expression rate were 9.09% (1/11), 46.67% (7/15) and 72.22% (26/36); gastrin The receptor positive expression rate of 0% (0/11), 53.33% (8/15) and 61.11% (22/36). Gastrin and gastrin receptor in gastric carcinoma total of 22 cases of positive and negative total 10 cases, the positive expression rate was positively correlated (P <0.05). Gastrin and gastrin receptor co-expression with gastric adenocarcinoma degree of differentiation, depth of infiltration, lymph node metastasis and NM stage (P <0.05). 2. Reg Ⅰ gene expression by RT-PCR in gastric carcinoma is strong, the positive rate was 69.44% (25/36); adjacent tissues, the positive rate was 40.00% (6/15); normal gastric mucosa, the positive expression rate of 18.18 (2/11)%. Gastric cancer specimens, gastrin and Reg Ⅰ gene co-positive 23 cases, a total of eight cases negative; the gastrin receptor Reg gene co-positive 20 cases, a total of nine cases of negative. Gastric adenocarcinoma between gastrin and gastrin receptor Reg Ⅰ gene positive expression rate was positively correlated (P <0.05). Gastrin and gastrin receptor Reg Ⅰ gene co-expression with the degree of differentiation of gastric adenocarcinoma, the degree of infiltration, lymph node metastasis and TNM stage (P <0.05). Reg Ⅰ and β-actin amplified fragment of 557 bp and 385 bp, respectively. Reg Ⅰ / β-actin was amplified bands grayscale ratio (Reg Ⅰ / β-actin) the mean value indicates the relative expression level of target gene (three times repeated experiments of (?) ± s said), the experimental group and the control group Reg Ⅰ / β-actin ratio of 0.54 ± 0.29 and 1.04 ± 0.22, respectively, both have a significant difference (p <0.01). Conclusion 1. Gastric tissue, gastrin and gastrin receptor highly expressed in performance between the two is a positive correlation. Gastrin and gastrin receptor co-expression of the degree of tumor differentiation, depth of invasion, lymph node metastasis, and TNM stage was significantly associated. 2. Gastric cancer tissues, Reg Ⅰ gene gastrin and gastrin receptor expression showed a positive correlation with the degree of tumor differentiation, depth of invasion, lymph node metastasis and TNM stage were significantly associated. 3. Inhibition of the expression of gastrin, gastric cancer cell line BGC-823 Reg Ⅰ gene expression was significantly inhibited. Reg Ⅰ gene gastrin promote gastric cancer development in a key downstream genes.
