Abstract

Typhoid fever is an acute systemic infection, including typhoid fever caused by Salmonella typhi (Salmonella typhi) and the A, B, paratyphoid Salmonella (Salmonella paratyphi A, B and C) caused paratyphoid. Paratyphi Salmonella paratyphi pathogens transmitted by the fecal - oral route, the infectious dose of 10 ~ 3 ~ 10 ~ 9, is the only host. The crowd is generally susceptible to Salmonella paratyphi, the highest of the children and young adults. It is estimated that each year in patients with the global typhoid (including paratyphoid) about 22 million, more than 20 million deaths. Since 1996, China paratyphi incidence increased year by year. More severe clinical drug-resistant strains of Salmonella paratyphi and multi-drug resistant strains (MDR) has a wide range of clinical treatment more difficult, some patients become carriers due to inadequate treatment. Therefore, we need to come up with a safe, efficient, simple, economic paratyphoid A vaccine to prevent. Immune proteomics is a strong immunity protein, this branch of the proteome, promising to be an effective way of looking for drugs and vaccines target pathogen proteome by two-dimensional electrophoresis separation, and then the patients free serum identification (because proof of these proteins can cause a host immune response). Salmonella paratyphi no capsule of Gram-negative bacteria, and its outer membrane protein has a very important role in the the pathogenic and stimulate the body's immune response aspects. Because the bacterial membrane proteins (in particular, outer membrane protein) antigen, often some of the reactive antigens, they have a very close interaction with the immune system. Therefore, the surface protein of the pathogen has a very important role in vaccine research. The experimentally the Salmonella paratyphi the outer membrane protein paratyphi Salmonella outer membrane protein two-dimensional electrophoresis reference map. Coomassie blue staining gel to take a total of 80 outer membrane proteins, 61 were identified, representing 30 proteins. Combined in accordance with the outer membrane protein of the genome prediction on the identification of all the isoelectric point of the protein spots / MW anastomosis extent, distribution and functional classification conducted a preliminary analysis. Further the immune proteome approach, bacterial outer membrane protein of the extracted two-dimensional electrophoresis separation, and then transferred to a PVDF membrane and immunoblotting reactions carried out with the the paratyphi patient convalescent serum, recording the adventitia of the immune response protein spots. Coomassie blue staining gel take outer membrane proteins and the immune response proteins digested MALDI-TOF mass spectrometry identification. 45 immune response in PVDF membrane, to 42 points were identified, representing 23 kinds of protein antigens, software analysis psort (www.psort.org), 13 kinds of outer membrane proteins, cytoplasmic proteins, the positioning is not clear (which may be multiple positioning) 1 species, unknown position 6. These proteins wish to become candidates for drug targets or vaccine ingredients.
