Abstract


Objective: To investigate the changes characteristic of CD4 CD25 regulatory T cells (CD4 CD25 regulatory T cell, Treg) in peripheral blood of gastric cancer before and after surgery, and to understand the role of Treg in tumor immunity. Methods: 38 patients with gastric cancer before surgery 14 days 3 days after surgery peripheral blood as an object of study, while collecting peripheral blood of 20 healthy persons as normal control group, the use of flow cytometry in their peripheral blood CD4 CD25 Treg, CD4 T and CD8 T-cell ratio of detection, the detection results with clinical and pathological parameters were statistically analyzed. Results: 1. Gastric cancer patients with preoperative peripheral Treg proportion was significantly higher than that of the control group and was statistically significant (P lt; 0.01). Comparison of peripheral Treg proportion of the control group after surgery, there was no statistical significance (P gt; 0.05). Peripheral Treg significantly higher proportion of postoperative proportion, with a statistically significant (P lt; 0.01) before surgery. Gastric cancer patients with preoperative peripheral blood CD4 T-cell ratio with the control group, no statistical significance (P gt; 0.05). Postoperative peripheral blood CD4 T cell ratio and the control group, the comparison between the two was not statistically significant (P> 0.05). Postoperative peripheral blood CD4 T-cell ratio compared with the preoperative, between statistical significance (P gt; 0.05). 3. Gastric cancer patients with preoperative peripheral blood CD8 T cell ratio was significantly lower than the control group, a statistically significant (P lt; 0.01). Postoperative peripheral blood CD8 T cell ratio with the control group, the comparison between the two was not statistically significant (P gt; 0.05). Postoperative peripheral blood CD8 T cells was significantly higher than the preoperative statistically significant (P lt; 0.01). Preoperative peripheral blood CD8 T-cell ratio and the proportion of Treg was negatively correlated (P lt; 0.05) 4. Peripheral Treg proportion of patients with gastric cancer before surgery in a statistically significant difference between the different lymph node metastasis and TNM staging (P lt ; 0.05), with lymph node metastasis and TNM stage were positively correlated (P lt; 0.01), with the depth of tumor invasion, distant metastasis, tumor differentiation and pathological types were not correlated (P gt; 0.05). 5 Preoperative and postoperative peripheral Treg proportion to the decline in the extent and depth of invasion, lymph node metastasis, distant metastasis, TNM stage, degree of differentiation, pathological type and surgical approach were not related to sex (P gt; 0.05). Conclusion:. Peripheral blood CD4 CD25 Treg proportion of patients with gastric cancer before surgery significantly increased, suggesting that it may be one of the mechanisms of tumor immune function in patients with suppressed, effective regulation and control of the group to the number of cells may contribute to tumor immune suppression status to correct and improve the therapeutic effect. 2 patients with gastric cancer before surgery in peripheral blood CD8 T-cell ratio decreased significantly, and its proportion of Treg proportion was negatively correlated, suggesting that Treg by inhibiting the proliferation and function of CD8 T cells and promote tumor cell growth and development. 3 patients with gastric cancer before surgery, peripheral Treg proportion with lymph node metastasis and TNM staging positive relationship, suggesting that Treg may be associated with tumor invasion, lymph node metastasis, Treg immunological parameters as the detection of disease progression to a certain extent. 4. Treg proportion of patients with gastric cancer after surgery compared with the preoperative decline, and the decline in the extent and depth of invasion, lymph node metastasis, distant metastasis, TNM stage, degree of differentiation, pathological type and surgical correlation, suggesting as long as the removal of the tumor itself peripheral blood, you can make the decline in the proportion of Treg, improve the body's anti-tumor immunity.
