Objective: To establish the immunosuppressive magnetization sorting technology and real-time quantitative reverse transcription polymerase chain reaction method of quantitative detection of circulating tumor cells, and to explore the clinical significance of quantitative detection of colorectal cancer in peripheral blood of patients with tumor cells. Methods: In vitro cultured colon cancer HCT-116 cells, will be divided into 101-106 of six different quantity levels, peripheral venous blood mixed with 5 ml of health, by density gradient centrifugation of peripheral blood mononuclear cells. Separated cells were divided into immune Enrichment group (A) and non-enriched group (B), another set of negative control group. Group A and the corresponding negative control group and package monoclonal antibody to CD326 immunomagnetic bead separation and enrichment of the role magnetic activated cell sorting device, extraction of total RNA enriched sorting specimens. Group B and the corresponding negative control group without immune enrichment processing, direct extraction of total RNA. Conclusion: The experimental study established the immunosuppressive magnetization sorting technique and RT-PCR detection method, the detection sensitivity is better than simply using RT-PCR detection method for the quantitative detection of tumor cells in peripheral blood by measuring the expression levels of hTERT-mRNA of . Colorectal cancer in peripheral blood of patients with hTERT-mRNA expression correlated with lymph node metastasis and TNM stage was significantly associated micrometastases in patients with colorectal cancer provide new ideas for early clinical. Further research, diagnosis, treatment and prognosis of patients with colorectal cancer have a greater clinical value.
