Abstract


Species of wild papaya wild papaya (Lardizabalaceae) Lardizabalaceae Latin scientific name Stauntonia chinensis DC, is an important development and medicinal value plants Herbs. It is commonly used as a traditional Chinese medicine, cardiac diuretic, analgesic efficacy of insecticide, Modern medical research has found that the main pharmacological effects is an anti-inflammatory analgesic block nerve conduction and radiation sensitizer, it is widely used in clinical the treatment of rheumatoid arthritis and other diseases. The systematic study of its analgesic effect of the extract and its mechanism is not yet reported, so this study on the basis of animal models to establish a variety of experimental pain study wild papaya rhizome extract (YMG) YMG aqueous phase extract (YMGW) and alcohol extract (YMGB) and its sub-fraction (YMGB1, YMGB2, YMGB3 YMGB4, YMGB5 YMGB6 and YMGB7) analgesic effect, and to explore the possible mechanism of effective fraction secondary development of of wild papaya botanicals and more in-depth research to provide the basis of scientific experiments. Now the research methods and results of the experiment are summarized as follows: The study first the three analgesia experiments carried out on the a wild papaya series extract the analgesic activity screening. Mouse acetic acid writhing test results show that: Compared with the control group, given YMGW, YMGB, YMGB1, YMGB3, YMGB4, YMGB6 YMGB7 (500 mg / kg, ig) were able to significantly inhibit the number of writhing, has good the analgesic effect; of wild papaya water extract YMGW and Stauntoniae alcohol extract YMGB and mouse hot plate induced pain in a dose of 500 mg / kg YMGB6 model had no analgesic effect; rat formalin inflammatory pain model, of YMG, YMGW, YMGB1 YMGB3 YMGB5 (400 mg / kg, ig) has a better analgesic effect, YMGB6, and YMGB7 has significant anti-inflammatory effect. Collectively, these results can be seen: the wild papaya extract fractions (YMG, YMGW, YMGB YMGB1-7) In addition to YMGB2 other fractions better analgesic effect. Secondly, the use of the mouse acetic acid writhing model and the rat formalin inflammatory pain model further evaluation of the relationship between dose and time-dependent on both models performed better analgesic anti-inflammatory effects of fractions. The results show that the YMG YMGB1, YMGB4 YMGB6 and YMGB7 five kinds of fractions in the concentration range of 320-976 mg / kg, with the increase of the dose, which induced by acetic acid in mice twisted body reaction inhibition increased gradually analgesic effect of the dose-dependent; YMGB5 YMGB6 and YMGW in the dose range of 100-800 mg / kg as the dose increased, the formalin-induced rat paw reaction and toe swelling degree of inhibition gradually increased with a dose-dependent analgesic anti-inflammatory effects. The results showed that the wild papaya water extract YMGW and ethanol extracts a good YMGB5 the analgesic effect may be achieved by reducing the expression of COX-2 levels and inhibition of enzyme activity; YMGB, YMGB4, YMGB6 and YMGB7 the analgesic effect may is achieved by inhibiting the COX-2 enzyme activity, thereby inhibiting PGE 2 production and release. The above results suggest that wild papaya extract fractions (YMG, YMGW, YMGB YMGB1-7) In addition to YMGB2 other fractions peripheral chemical stimulation pain certain extent, central heat stimulation analgesic effect not significant; in a dose-dependent analgesic anti-inflammatory effects, effective analgesia time is about one hour; the effective fraction Stauntoniae analgesic effects and opioid receptor analgesic pathways unrelated, primarily by inhibiting the activity of COX-2 2 of the formation and release of PGE and expression, thereby inhibiting play outer peripheral analgesic action.
