Abstract


This paper first reviews biological alternatives as synthetic plastics biodegradable plastic poly-β- hydroxybutyrate (PHB) , the nature of physiological functions and applications , as well as PHB the four production pathway , and focuses reported microbial fermentation production of PHB Research Survey , as well as microbial intracellular PHB the extraction and PHB analysis method research progress . The Sphaerotilus filamentous bacteria in the activated sludge , has a strong ability to degrade organic matter in sewage and toxic substances , and intracellular able to accumulate a considerable amount of PHB , has gradually taken seriously , therefore , the paper reported natans Research Survey , and the research direction in the strain of PHB , to carry out the basic research of the system mainly include the following: 1 . Isolated by enrichment culture and crossed combined screening and rescreening means . , Successfully separated from the activated sludge to a high yield of PHB jersey bacteria ( Sphaerotilus natans ) . 2. Berger bacterial identification manual cell morphology and structure , culture characteristics and physiological and biochemical characteristics of the strains were identified , and the Culture Collection methods preliminary study . 3. Strains producing PHB fermentation optimization studies, preliminary to determine the optimum fermentation medium and fermentation conditions . 4. Adopt different methods to study of the Sphaerotilus intracellular PHB extract , initially identified the best extraction method . 5. PHB samples using the extract obtained in the ultraviolet absorption spectroscopy ( UV ) , infrared spectroscopy ( IR ) and elemental analysis , nuclear magnetic resonance (NMR) and gas chromatography (GC) , etc. carried out by means of qualitative and quantitative analysis . The above research will Sphaerotilus purify sewage and fermentation production of PHB to provide a theoretical basis for the future development and utilization .
