The purpose of the establishment of the fluorescent quantitative RT-PCR analysis of the WT1 gene , WT1 gene expression in various types of leukemia , and the significance of the detection of minimal residual disease . Method RT-PCR amplification of WT1 gene in K562 cells and β-actin gene , gel cutting and recycling and purification , as a standard template after UV spectrophotometer quantitative . Fluorescent quantitative RT-PCR method detected 80 cases of leukemia in patients with bone marrow or peripheral blood and 10 cases of normal human peripheral blood WT1 gene expression , and long-term tracking of WT1 gene expression in 15 patients with acute leukemia . The experimental data were statistically analyzed using spss software . Results of 10 patients with newly diagnosed acute leukemia patients with bone marrow and peripheral WT1 gene expression was no significant difference . Leukemia patient group ( 49 cases of AML , 18 patients with ALL , 7 cases of HAL, 6 CML ) and normal Shanxi Medical University, a master's degree thesis the peripheral blood WTI gene expression were significantly different expression of myeloid than lymphoid , MS significantly low expressed. WTI gene expression and gender , the incidence of white blood cell count , the karyotype was no significant correlation . The high levels of wTI expression CR rate in patients with long-term survival of the short , the prognosis is poor . On clever peripheral blood of patients with acute leukemia WTI gene expression tracking long-term testing, four cases had reached complete remission in patients with 3 7 . Months after recurrence , two cases before clinical relapse February 1 , WTI levels significantly higher , by at least one pairs number of levels . Three cases no significant changes induced resistance to chemotherapy in patients with WTI level always . Conclusion fluorescence quantitative RT-PCR was used to detect the WTI gene high sensitivity and good specificity . Compared with normal peripheral blood , WTI was highly expressed in all types of leukemia and conditions related to expression level and disease . The fluorescence quantitative RJ'-PCR method WTI quantitative analysis of gene expression in peripheral blood of patients with acute leukemia , can provide a useful tool for monitoring minimal residual disease .
