Seven flower (Heptacodium miconioides) under the the Caprifoliaceae (Caprifoliaceae) Seven genus (Heptacodium) the single species of plant, only distributed in a few areas such as Zhejiang, Anhui, has been identified as the two national key protected plants. Use the ISSR and cpSSR two molecular markers to explore of the Population genetic diversity and genetic differentiation and population spatial genetic structure, reveals Seven flower endangered mechanism and develop appropriate protection measures. The results are as follows: 1. Using ISSR Markers located in the roof of five of the Population genetic diversity and genetic differentiation. The Population genetic diversity of 58.17% at a medium level. A variety of genetic diversity Lion Rock pit population (41.18%), the Zhe Village Population lowest (33.33%). Coefficient of genetic differentiation (GST) was 0.3497, more obvious genetic differentiation between populations. Genetic variation within populations (64.59%). Gene flow between the various groups of 0.9299, indicating that the seven flower gene flow is somewhat limited. The clustering results the East Gate Ridge and the hills were clustered together, philosophy Village, Lion Rock pit and longitude hole were clustered together. The heterogeneity of the microenvironment may be the cause of forming such a genetic characteristics. 2 using cpSSR marker analysis cpDNA genetic diversity, the Seven spent cpDNA not detected on the genetic variation, the findings of the project to verify the conclusions of scholars in the past, and speculated that may be left after the Ice Age only seven flower mother plant populations . By ISSR Marker spent on the seven small-scale spatial genetic structure study designed to investigate the the Seven Flower distance and genetic distance correlation. Of the Population in the spatial distance from 0 to 8.06m, all plants showed significant spatial genetic structure, ie aggregated distribution, the rest of the distance, spatial genetic structure is not significant. Plants of different age class-level tree I showed aggregated distribution pattern of spatial scales range from 0 to 4.36m class II tree of 0 to 6.42M III level tree to 3.3m tree level IV of 0 to 8.06m. With the lowering of the age of a tree, showing significant spatial genetic structure increasing the distance, the more similar genotype together, gene flow is restricted. The cause of this spatial pattern may be due to the limited spread of the pollen, micro-environmental heterogeneity and inbreeding depression. Seven Populations of genetic diversity in the middle level, a variety of genetic diversity, which may be one of the reasons that lead to the seven flower endangered. From the spatial genetic structure of the populations, gene flow between the Population hampered, leading to resist the external environment changes reduced genetic variation, so that the population tends to be increasingly endangered. Combined with the results of previous studies, Seven spent on protective measures: step up publicity to reduce vandalism, to expand the size of the population, and expand mate expand the gene flow between populations, and actively carry out the study of the breeding ecology, in situ and ex situ conservation.
