Abstract


Denervated striatum proteomic analysis of the experiment in the first part of the Parkinson's disease model 6 - hydroxydopamine Parkinson's disease model denervated striatal proteome expression changes were studied. We found to extract promote the in vitro culture of embryonic ventral midbrain dopaminergic neuron survival of dopaminergic neurons innervating the striatum. In order to further study the molecular basis of the nutritional role, looking for a new promotion of dopaminergic neurons damage repair factor, in this study, the comparative analysis of two-phase gel electrophoresis proteome expression changes in the striatum after denervation. The results showed no significant difference in the vast majority of protein expression levels in the striatum after denervation, but a small number of protein expression levels changed significantly: 7 protein expression increased 4 protein expression decreased. After matrix-assisted laser desorption ionization - time of flight mass spectrometry to determine which eight types of protein, and protein expression levels increase include: amyloid precursor protein (amyloid precursor-like protein 2 APLP2), bradykinin original (kininogen) Glucokinase (glucokinase), brain-derived type α tropomyosin (tropomyosin αchain type brain-1, TMBR-1), in accordance with the level of expression of troponin I light chain (calpactin Ⅰ light chain, Calpactin Ⅰ-lc); downward a protein comprising: a neural epidermal growth factor-like protein 2 (Neural epidermal growth factor-like 2, Nell 2), thyroid hormone receptor beta-2 (of the THYROID hormone receptorβ-2 the THRβ-2), small chromosome maintained factor 6 (Mini -chromosome maintenance 6, MCM6). Immunohistochemical staining showed that dopaminergic neurons of the substantia nigra co-expression of APLP2 and tyrosine hydroxylase. 6 - OHDA lesions in the nigrostriatal pathway, APLP2 expression in the substantia nigra compacta With the death of dopaminergic neurons decreased in the striatum and substantia nigra pars reticulata with denervation APLP2 Expression rise. Moreover, over-expression of the chondroitin sulfate-modified APLP2 (APLP2-751) Chinese hamster ovary cell line conditioned medium promote vitro embryonic ventral midbrain dopamine neurons survive overexpression without chondroitin sulfate-modified APLP2 (APLP2 -763), Chinese hamster ovary cell line conditioned medium does not have the same role. These results suggest that chondroitin sulfate-modified the APLP2 substantia nigra dopaminergic neurons potentially lt; WP = 6 gt; protect and promote the role of damage repair.
