With the development of modern medical technology and materials sciencedevelopment, especially, the emerging area of inorganic nanoparticles entrappingbiomolecules has already exhibited its diversity and potential applications in manyfrontiers of modern material science. Some of the novel inorganic carriers for genedrug delivery have been a hot spot in the ifeld of medicine transportation in recentyears such as gold nanoparticles, silica nanoparticles, calcium phosphate nanoparticles,manganous phosphate nanoparticles and other inorganic phosphate nanoparticles. Thecalcium phosphate nanoparticles could translocated into cells because it had goodbiocompatibility,bioabsorbability and biodegradability, lack toxicity and stablechemical properties, on the other hand,it also could be exploited to deliver a broadrange of therapeutics including proteins, antibodies, oligonucleotides, imaging agentsand liposomes in a variety of situations and biological systems. Calcium ions ofcalcium phosphate carirer can form chemical bond with phosphate backbone groupsof antisense oligonucleotides (ASODNs), it’s one of the eiffcient methods forprotecting ASODNs transfer into cell lines,drug carirer loaded antisense drugs can beselectively concentrated in cancerous site to reduce drug degradation and losses,reduce drug cytotoxicity to normal cells or tissue and improve drug availability.In this paper, we prepared with Mn doped-nanoscale spherical phosphate as drugcarrier. The appropriate three-phase system was choosed to control the size ofnanoparticles by the microemulsion method. The nanoparticles doped manganese hasa good effect of MRI which could get visual observation in the cell toxicity anddistirbution. We build a new type of antitumor drug system and new drug useefficiency provides research foundation. Finally, We obtained the fluorescent markersof anti-cancer drugs and imaging function, for the development of multifunctionaldrug provides research foundation. This paper includes two part contents as follows:1, The ifrst part content mainly discusses preparation of nanometer hydroxyapatite with aqueous phase method. The results show that it has succeed for preparingnanoscale spherical Mn doped-hydroxyapatite nanoparticles. In vitro cytotoxicitystudies of hydroxyapatite nanoparticles using the MTT assay with Bxpc cells tocytotoxicity, the results show Mn doped-hydroxyapatite have a good biological safetyin a certain concentration range of concentration.2. The first part content mainly discusses the construction of the Mn doped?phosphate drug-loading system and application in the biology. Then, appropriatecalcium phosphate nanometer size and concentration of Mn doped-drug carrier waschossed and the nanoparticles with the fluorescence properties and targeted featureswere made through a series of modifietion. Nanoscale Mn doped-spherical phosphateswere successfully synthesized by reversed-phase microemulsion method. Wemodiifed the nanoparticles with mPEG-NHyThen, the nanoparticle and targetmolecules MAB-C225were joined together by glutaraldehyde crosslinking reaction.Atfer the the adsorption of DOX, the nano drugs carirer which could get the visuallyobserve toxicity and distirbution in cells was well fabricated. The effect of MRIimaging of biological phenomena of the drug carrier which was applied to biologicalenhanced molecular magnetic resonance image signals in the body were studied.
