Objective:The etiolog and mechanism of Intracranial aneurysms information is not clearRecently some articles reported hemodynamic changes in endothelial damages andhemal wall inflammation may be tumor-initiating link in intracranial aneurysmsinformation.VE-cadherin-mediated adhesions are important for the control ofEndothelial injury repair, of Maintain vascular permeability,of leukocyte extrav-asation and Intracellular signal transduction. We have recently found that, inintracranial aneurysms wall, the expression of VE-cadherin significantly reduced,and on the basis of both at home and abroad to intracranial aneurysm formationmechanism and vascular endothelial cells and its regulation connections between thetwo aspects of the research progress, and application experimental rat intracranialaneurysms in the model simulation intracranial aneurysms in the process of theformation of blood vessel walls damage, the aneurysm formation process of SRCkinase and VE-cadherin phosphorylation level expression.Methods:1.Sixty male rats of Sprague-Dawley were randomly allocated into2groups:silver clip group,of which40rats were constricted both renal arteries with two silverclips of an inner diameter of0.30mm and ligated left common carotid artery; then,the silver clip group were fed with2%saline after procedures; and control group,ofwhich20rats were served with only sham operation. Postoperative normal feed.Threemonths after the procedure, the rats were killed. The formation of intracranialaneurysms and their pathological changes were observed by lightmicroscope.Aneurysmal changes can be found in the junction of the right anterior cerebral (ACA)and the olfactort artery (OA) of rats.2. Applications of molecularly protein imprinting detect the expression of SRCkinase and phosphor-VE-cadherin in rats intracranial aneurysm, and Applications ofreal-time quantitive PCR detect the expression of SRC kinase in rats intracranialaneurysm Results:1. The protein imprinting results: The experimental group SRC kinase、phosphor-VE-cadherin (Tyr-685)、 phosphor-VE-cadherin (Tyr-658) andphosphor-VE-cadherin (Tyr-731) have obvious expression.In the contrast of normalgroup, SRC kinase、 phosphor-VE-cadherin (Tyr-685)、 phosphor-VE-cadherin(Tyr-658) and phosphor-VE-cadherin (Tyr-731) expression significantly increased2. The real-time quantitive PCR results: In the contrast of normal group, SRCkinase mRNA expression increased in the experimental groupConclusion:SRC kinase、phosphor-VE-cadherin (Tyr-685)、phosphor-VE-cadherin (Tyr-658)and phosphor-VE-cadherin (Tyr-731) was significantly overexpressed in experimentaneurysms than the normal control group of expression, our experimental resultspreliminarily show that SRC kinase, and VE-cadherin phosphorylation expressionmay be involved in the formation of intracranial aneurysms.
