ObjectiveTo investigate the expression levels and locations of two proteins, JAM-1 and claudin-7, and their mRNA levels in skin lesions from patients with psoriasis vulgaris and explore the possible roles of JAM-1 and claudin-7 in the pathogenesis of psoriasis vulgaris.Methods25 cases of patients with psoriasis vulgaris and 15 cases of healthy persons were selected.1) to detect the expression levels and locations of JAM-1 and claudin-7 proteins in skin lesions of patients with psoriasis vulgaris and normal skin tissues by immunohistochemistry method;2) to determine the mRNA expression of JAM-land claudin-7 in skin lesions from patients with psoriasis vulgaris and normal controls by reverse transcription polymerase chain reaction (RT-PCR).Results1) Compared to normal skin tissues, there were no JAM-1 and claudin-7 expression in the lesions from patients with psoriasis vulgaris.2) The mRNA expression of JAM-1 in psoriatic skin lesions were significantly lower than that in normal skin tissues (0.38±0.10 vs 2.51±0.09). The mRNA expression of claudin-7 in psoriatic skin lesions were significantly lower than that in normal skin tissues (0.27±0.12 vs 1.83± 0.15).ConclusionThe proteins of JAM-1 and claudin-7 are negatively expressed in skin lesions from patients with psoriasis vulgaris, indicating that skin barrier is impaired in psoriasis vulgaris.
