Objective To detect the protein and mRNA expressions of CXCL12 and CXCR4 in endometrial carcinoma, and to evaluate their roles in carcinogenesis and metastasis.Methods 1. Specimens of endometrial carcinoma, atypical hyperplasia endomembrane, simple hyperplasia endomembrane and normal endometrial tissue were collected and processed into tissue chips. The protein expressions of chemokines CXCL12 and its receptor CXCR4 in the samples were analyzed by immunohistochemistry; 2. The mRNA expressions of chemokines CXCL12 and its receptor CXCR4 in the endometrial carcinoma and normal endometrial tissue were analyzed by RT-PCR. The relationship between the protein and mRNA expressions of chemokines CXCL12 and its receptor CXCR4 in endometrial carcinoma and FIGO stage, metastasis of lymph nodes, the degree of differentiation, the type of pathology, and the age of patient were analyzed. The results of this study were analyzed SPSS 13.0. The significant size of test was validated as value of 0.05.Results The protein positive expression of CXCL12 in endometrial carcinoma was higher than those in atypical hyperplasia endomembrane, simple hyperplasia endomembrane and normal endometrial tissue(P<0.05、P<0.05、P<0.05). The differences of CXCR4 protein positive expression among the endometrial carcinoma、atypical hyperplasia endomembrane, simple hyperplasia endomembrane and normal endometrial tissue were significant(P<0.05)o Stronger expression of CXCL12 mRNA was observed in endometrial carcinoma, compared with that in normal endometrial tissue(P<O.05). The protein and mRNA expressions of CXCL12 was not associated with FIGO stage, the degree of differentiation, the type of pathology, lymph node metastasis and the age of patient (P>0.05). The protein and mRNA expressions of CXCR4 was closely related to the degree of differentiation (P<0.05), but it was not associated with FIGO stage, the type of pathology,lymph node metastasis and and the age of patient (P>0.05). The protein and mRNA expressions of CXCL12 was positively correlated with CXCR4 in endometrial carcinoma (r=0.426 and 0.796, P<0.01).Conclusion The high-level expression of CXCL12/CXCR4 implied that the interactions between CXCL12 and CXCR4 play important roles in the carcinogenesis and progression of human endometrial carcinoma.
