Expression and Significance of COX-2 Protein and Bcl-2 Protein in Distal Transitional Mucosa Adjacent to Rectal Carcinoma

Abstract


Objective:To investigate the expression of COX-2 protein and BCL-2 protein in transitional mucosa adjacent to rectal carcinoma, and judge the transitional mucosa whether the cancer precursor event.Methods:Mucin histochemical method(HID/AB)was employed to determine the distal mucosa 2cm away from rectal carcinoma and find the transitional mucosa in 54 cases of rectal carcinoma. Immunohistochemical methods was employed to detect the expression of Bcl-2 and COX-2 protein in the specimen of rectal carcinoma mass, transitional mucosa, non-transitional mucosa, and 20 cases of normal rectal mucosa, and count the scores of the expression of COX-2 protein and Bcl-2 protein. T and chi-square test were preformed for data analysis.Results:19 cases of positive TM were found in all the distal mucosa 2cm away from rectal carcinoma. The positive expressions of COX-2 and Bcl-2 protein can be found in carcinoma mass and distal mucosa. COX-2 expression was recorded in 81.48% of carcinoma mass,21.05% in transitional mucosa,17.14% in non-transitional mucosa,10% in normal mucosa. Bcl-2 protein expression was recorded in 77.78% of carcinoma mass, 21.05% in transitional mucosa,22.86% in non-transitional mucosa,5% in normal mucosa. For the COX-2 and Bcl-2, intensity and distribution were classified and scored. Significant contrast was observed for carcinoma mass and transitional mucosa (p<0.01). But there were no significant contrast between the transitional mucosa and non-transitional mucosa (p>0.05), transitional mucosa and normal mucosa(p>0.05).Conclusions:The performance of transitional mucosa was not the cancer precursor event, but the non-specificity reaction of carcinoma or inflammation.
