Abstract

Objective: This experimental study is based on network theory of governance, application security, reasonable and effective dose Huoxuetongluo traditional Chinese medicine to improve rabbits with acute myocardial infarction, revascularization, myocardial no-reflow Organization myocardial ultrastructure and enzymatic expression, molecular studies of myocardial microcirculation lead to coronary heart disease mechanism and a preliminary study, research data provide a basis for clinical myocardial protection. Materials and Methods: 50 adult of New Zealand white rabbits were randomly divided into 5 groups (n = 10), ligation of the rabbit left ventricular branch 90min, release the ligature infusion 120min production of acute myocardial ischemia-reperfusion injury model; since the beginning of reperfusion, ② group from the marginal ear vein with normal saline (based upon 0.3ml · kg-1 · min-1 speed continuous intravenous infusion); (3) (4) group at the dose given ginaton (first 2mg · kg- intravenous injection, then press 0.3mg · kg-1 · min-1 speed continuous intravenous infusion); the ⑤ group was given nitroglycerin (0.1 · kg-1 · min-1 speed continuous intravenous infusion). To reperfusion 120min discontinuation of the infusion. Collecting blood samples of serum creatine kinase (CK), lactate dehydrogenase (LHD) leakage rate from right at the end of the experiment room at the right atrial appendage after myocardial tissue by reverse transcription polymerase chain reaction (RT-PCR) The method for determination of myocardial tissue cPLA2-gamma gene expression levels, take another left ventricular apical myocardial tissue morphology was observed by electron microscopy of myocardial ultrastructure. Results: Compared with the sham group, model group with ginaton A, group B, the nitroglycerin group of cPLA2 mRNA expression was significantly higher (P lt; 0.01), ginaton A group, B group cPLA2 mRNA expression increased in the normal group, but compared with the model group was significantly lower than the model group (P lt; 0.01). Nitroglycerin cPLA2 mRNA expression increased in the normal group, but compared with the model group was significantly lower than the model group (P lt; 0.01). Ginaton A B group and nitroglycerin group showed no significant difference. Myofibrillar fracture compared with sham group, model group, dissolved, disappeared blank area, swelling of the sarcoplasmic reticulum, mitochondria (Mi), the outer membrane is incomplete Mi myelin degeneration, glycogen disappeared, intercalated disc swelling. Compared with the model group Kim Nadeau treatment group the myofibrillar neat sarcomere obvious Z line clear, an increase in the number of sarcoplasmic reticulum Mi, increased glycogen granules, intercalated disc close to normal. 3. Jinnaduoming significantly reduced serum CK, LDH activity. Conclusion: 1. Ginaton improve rabbit myocardial ischemia-reperfusion injury systolic function, reduce cPLA2-gamma gene expression, reduce enzyme leakage rate, reducing myocardial ultrastructural damage. 2. Ginaton on myocardial ischemia-reperfusion injury in rats, and its mechanism may inhibit the generation of oxygen free radicals and inhibit lipid peroxidation.
