Abstract


Purpose: The purpose of this experimental study is to detect Bag-1 gene (Bcl-2 binding anti-apoptotic gene 1) in colorectal cancer tissues and normal colorectal tissue changes and the relationship between lymph node metastasis; investigate Bag-1 and biological behavior of colorectal cancer, further study of colorectal cancer development mechanism for colorectal cancer diagnosis, treatment and prognosis of patients judged to provide newer and more effective method. Methods: protein immunoblotting (Western blot) and reverse transcription - polymerase chain reaction (RT-PCR) method combined detection after surgical removal of the 55 cases of pathologically confirmed colorectal cancer tissue samples Bag-1 expression in the control group was 25 cases of normal colorectal mucosa tissues (such as: trauma patients). Results: (1) protein immunoblotting (Western Blot) results showed that: 55 cases of colorectal cancer were detected in different levels of Bag-1 protein expression by Western blot analysis software Optimas 6.0 in all cases the protein bands were analyzed, it was found , Bag-1 protein in primary tumor tissue protein with a relative intensity of each case (5 ~ 109,64.09 ± 31.59); lymph node metastasis in colorectal cancer tissue protein relative intensity (30.34 ~ 109,81.05 ± 19.64) than without lymph node metastasis of colorectal cancer protein relative intensity (5 ~ 70.13,26.19 ± 16.32) (P lt; 0.001), but both were significantly higher than the normal mucosa tissues Bag-1 protein relative intensity (0-10,4.28 ± 2.82) (P lt; 0.001), which all have significant difference. (2) reverse transcription - polymerase chain reaction (RT-PCR) showed that: Bag-1 and β-actin primers amplified gene fragment by electrophoresis and EB staining electrophoretic bands were clearly visible, size of the amplified fragment designed with exactly the same size, respectively, 331bp, 254bp. Software testing analysis, we found 55 cases of colorectal carcinoma Bag-1mRNA expression relative value (0.33 ~ 1.21,0.74 ± 0.26). Bag-1mRNA with lymph node metastasis in colorectal cancer tissue expression levels relative value (0.52 ~ 1.21,0.87 ± 0.21) than those without lymph node metastasis in colorectal cancer tissue expression levels relative value (0.33 ~ 0.65,0.45 ± 0.09) (P lt; 0.001), but both were significantly higher than the normal mucosa tissues Bag-1mRNA expression relative value (0 ~ 0.22,0.10 ± 0.06) (P lt; 0.001), which all have significant difference. Conclusion: Bag-1 overexpression in colorectal cancer occurrence and development plays an important role; Bag-1 expression in colorectal cancer lymph node metastasis, Bag-1 overexpression promotes lymph node metastasis of colorectal cancer, which can be used as predictive intestine carcinoma metastatic potential of biological indicators to detect the expression of Bag-1 levels may contribute to patient prognosis.
