Effects of All-trans Retinoic Acid on High Glucose-induced Expression of ACE/ACE2 in Human Renal Tubular Epithelial Cells

Objective To investigate the influence of all-trans retinoic acid(ATRA)on the expression of angiotensin converting enzymes(ACE),angiotensin converting enzymes2(ACE2)and AngiotensinⅡ(AngⅡ)in human renal tubular epithelial cells induced by high glucose and to probe the role of RAS in the pathogenesis of diabetic nephropathy(DN) and the intervention situation of ATRA.Methods HK-2 cells cultured in vitro were randomly divided into several groups:(1)normal glucose group(NG), high glucose group(HG) and mannitol group(MG,).Each group were cultured 24h,48h and 72h respectively;(2)HK-2 cells in HG group were exposed to medium containing 30mmol/L D-glucose and different concentrations of ATRA (ATRA1 group 10-7mol/L、ATRA2 group 10-6mol/L、ATRA3 group 10-5mol/L), or 10-2mol/L ethanol(ethanol group),or 10-5mol/L captopril(Captopril group).Each group were cultured for 48h. The expression of ACE and ACE2 mRNA in HK-2 cells were detected by reverse transcription polymerase chain reaction (RT-PCR) and the expression of ACE and ACE2 protein were detected by Western blot . The concentration of AngⅡin the culture medium was detected by enzyme linked immunosorbent assay(ELISA). Results (1)In comparison with NG group,the mRNA and protein expression of ACE in HG group increased significantly in a time dependent manner (P<0.01). The expression of ACE2 in mRNA and protein in HG group at 48 hours and 72 hours after induced by high glucose decreased significantly in comparison with NG group (P<0.01). The level of AngⅡin HG group were higher significantly than in NG group(P<0.05).(2)The expression of ACE mRNA and protein in HK-2 cells cultured under high glucose joining 10-7mol/LATRA were inhibited significantly in comparison with HG group (P <0.05), and the inhibition increased with the dose increasing,and this difference was statistical significance (P <0.01).While 10-7mol/LATRA began reversing the expression of ACE2 mRNA and protein in HK-2 cells induced by high glucose at 48h after joining ATRA (P <0.05), and the reversal enhanced with the dose increasing. The difference were statistical significance (P <0.01). The concentration of AngⅡin groups with adding 10-7mol/LATRA already decreased in compared with HG group (P <0.05),and the concentration of AngⅡin 10-6mol/LATRA group and 10-5mol/LATRA group decreased significantly in compared with NG group (P <0.01).Conclusion (1)High glucose can induce ACE expression and inhibit ACE2 expression ,then promote synthesis of AngⅡin proximal tubular cells(.2)ATRA can increase the expression of ACE2 and inhibit the expression of ACE, and then inhibit the expression of AngⅡinduced by high glucose in HK-2 cells,which may play an important role in delaying diabetic nephropathy.
