Background Obesity is associated with hypertension, type 2 diabetes and other chronic non-communicable diseases are closely related, and is widely popular on a global scale, the prevention and treatment of obesity has become one of the key aspects of the chronic disease prevention. Obesity is the result of the combined effects of genetic, environmental and other factors, more and more evidence to suggest that genetic factors play an important role in obesity related to energy consumption or energy intake gene may be associated with obesity, β2 adrenal hormone receptor (ADRB2) gene and uncoupling protein 2 (UCP2) genes are genes affect energy consumption. ADRB2 and UCP2 through different biological mechanisms affect the body's energy metabolism, their metabolic pathways mutual adjustment mechanism, suggesting that the combined effect that may exist between the two gene polymorphism may increase the probability of obesity phenotype multi inconsistent findings on ADRB2Arg16Gly and of UCP2-866G / A polymorphism with obesity relationship, and specific ADRB2Arg16Gly and UCP2-866G / A polymorphism the joint effects associated with obesity also less, not a large sample The study of natural populations. Select the Beijing Shijingshan Shanxi Yuxian two groups of natural populations to explore the relationship of these two loci polymorphism with obesity and their combined effect of behavioral factors affect the relationship of these genes and obesity. Objective To understand the situation of China's two places the natural populations ADRB2Arg16Gly sites and UCP2-866A / G polymorphism; explore these two sites whether polymorphisms in obesity-related phenotypic changes caused by the combined effect of; explore two loci polymorphism and behavioral factors jointly affect the occurrence of obesity. Methods 2004 and 2005, Shanxi Province of Yuxian crowd and Beijing Shijingshan crowd survey of cardiovascular risk factors. The subjects including Shanxi the Yuxian and Beijing Shijingshan 2 cohort, age 56 ± 10 years old. The completion of the on-site survey, a total of 3753 people, a response rate of 84%, which have ADRB2Arg16Gly locus polymorphism detection result by 3413, UCP2-866A / G polymorphism test results 3701, two-point results and other relevant information complete by 3365. Rule out cancer, heart attack and stroke after 3138 as the research object. Used questionnaires to collect demographic, medical history, and smoking, drinking, physical activity and other behavioral data. Salting-out method to extract DNA using PCR-RFLP method for genotyping, DNA MassARRAY technology to detect two-locus genotype. Use the SPSS13.0 package for statistical analysis, the mean of the group compared using t-test and variance analysis, using the general linear model analysis of the differences of the different genotypes and alleles between obesity indicators. The classification variable frequency distribution groups compared using the chi-square test, the use of non-conditional logistic regression model analysis dangerous differences in obesity prevalence between different genotypes, bilateral P lt; 0.05 as statistically significant standard. Results the the 1. ADRB2Arg16Gly sites and UCP2-866A / G locus distribution ADRB2Argl6Gly point AA, GG and AG genotype frequencies were 0.368,0.158 and 0.474, genotype frequencies conformed to Hardy-Weinberg balance (P gt; 0.05 ), male and female genotype frequency difference was not statistically significant (P = 0.618). UCP2-866A / G locus genotype frequencies of AA, GG and AG were 0.218,0.284 and 0.498, genotype frequencies in Hardy-Weinberg balance (P gt; 0.05), male and female genotype frequency differences were statistically significant (P = 0.009). Analysis of male and female sexual ADRB2Argl6Gly sites 2.β2 adrenergic receptor gene polymorphism is associated with obesity, obesity-related indicators and behavioral factors in the three genotypes, the difference was not statistically significant (P gt; 0.05), in men and women four The GLM model different covariates combinations were not found of different genotypes ADRB2Argl6Gly sites and different allele type between BMI and waist circumference difference was statistically significant (P gt; 0.05). Were not found in men and women Multivariate Logistic regression model the ADRB2Argl6Gly polymorphism with obesity and abdominal obesity is related to the occurrence (P gt; 0.05). 3 uncoupling protein 2 gene polymorphism with obesity associated analysis of men and women the UCP2-866A / G point three genotypes differences in obesity-related indicators and behavioral factors was not statistically significant (P gt; 0.05). GLM After adjustment for all covariates in the male control group (BMI ≥ 28kg/m2) AG genotype greater BMI (P = 0.032), a combination of four different covariates in male obesity and abdominal obesity group GLM model were found in the waist of the differences between the different genotypes of UCP2-866A / G locus statistical significance (P lt; 0.05), male and female differences were not found in the different genotypes and alleles between BMI. UCP2-866A / G polymorphism with obesity in men Multivariate Logistic regression model for (P = 0.019), but has nothing to do with abdominal obesity occurred (P gt; O.05), women did not find UCP2 The locus-866A / G polymorphism with obesity and abdominal obesity related to the occurrence of (P gt; 0.05). Analysis of male and female sexual β2 adrenergic receptor genes and uncoupling protein 2 gene in different combinations of genes 4.β2 adrenergic receptor genes and uncoupling protein 2 gene combined with variation and obesity associated obesity-related indicators and behavior of the difference was not statistically significant (P gt; 0.05). In the the GLM model portfolio by the men and women of four different covariates were not found between BMI and waist circumference in ADRB2Argl6Gly and UCP2-866A / G genotype and allele difference was statistically significant (P gt; 0.05). ADRB2Argl6Gly and UCP2-866A / G polymorphism of the combined effects of obesity and abdominal obesity were not found in men and women Multivariate Logistic regression model related to the occurrence (P gt; 0.05) 5. Multivariate Logistic regression of male and female were not found in the model the ADRB2Argl6Gly sites, sites of UCP2-866A / G, and ADRB2Argl6Gly and UCP2-866A / G polymorphism of the combined effects of physical activity, smoking and drinking there was an interaction would increase the risk of obesity and abdominal obesity (P gt; 0.05) Conclusion This study did not found the natural population ADRB2Argl6Gly site polymorphism associated with obesity. UCP2-866A / G polymorphism men found rates of obesity, and abdominal obesity rate; women not UCP2-866A / G polymorphism with obesity and abdominal obesity is related to the occurrence (P gt; 0.05). UCP2-866A / G locus genotypes were not found in men and women and between allele associated with BMI, but male obesity and abdominal obesity group were found between the different genotypes of UCP2-866A / G locus Waist The difference was statistically significant, suggesting that UCP2-866A / G polymorphism may be associated with the position of the male obese fat distribution. Not found ADRB2Arg16Gly of UCP2-866A / G and two points polymorphism and behavioral factors interaction in the presence of obesity and abdominal obesity.
