The purpose of identification of human breast normal tissue and cancer tissue, cancer tissue and human breast cancer cell line MCF-7 in the SP (side population cells, side population) cells content whether there are differences; compare human breast cancer cell line MCF-7 in, SP cells with NSP (non-side population cells, the non side population) characteristics of the cell surface markers, analysis the SP cells content with the relationship of the factors affecting the prognosis of breast cancer. Further explore the breast cancer stem cells. Method 1. Preparation of human breast normal tissue, cancer tissue, cancer tissue cell suspension and the human breast cancer cell line MCF-7 cell suspension joined Hoechst33342 staining in a group, another group at the same time adding verapamil by The flow cytometry SP cell content; 2 stars selected by flow cytometry in human breast cancer cell line MCF-7 in the SP cells and NSP cells, detection of these two cell surface protein or antigen and cell cycle proliferation conditions; 3. analysis of SP cells content on the prognosis of patients with breast cancer. Results 1.SP cells in normal human breast tissue, cancer tissue and cancer tissue and human breast cancer cell line MCF-7, the contents were (0.54 ± 0.17)% and (7.23 ± 1.12)%, (8.34 ± 0.96). % and (9.58 ± 1.27)%, and the content of SP cells between them with a significant difference (P lt; 0.05); 2.CD38, CD117 SP cell populations of CD34 in and NSP cell populations in the expression rate of no significant difference (P = 0.256, P = 0.920, P = 0.317); SP cell populations, CD133, of ABCG2 expression rate was significantly higher than the rate of the NSP cell population expression (P = 0.000, P = 0.000); CD24 The SP cell populations in the expression rate was significantly lower than the rate of the NSP cell populations expression (P = 0.000); 3.SP cell population, significantly higher than the proportion of cells in G0/G1 quiescent NSP cell populations (P = 0.000); while in the S, G2 / M phase of the cell number in the NSP cell populations significantly more than in the SP cell population (P = 0.001, P = 0.000); cells in the NSP cell populations in high expression of Ki-67, and the difference SP cells was significantly (P = 0.000); SP cells have a lower rate of apoptosis, the NSP cell apoptosis rate higher, both compared to the presence of a significant difference (P = 0.000); breast cancer patients, the content of the SP cells with the risk factors of breast cancer patients have a significant correlation (P lt; 0.05). Conclusions normal human breast tissue, cancer tissue and cancer tissue and human breast cancer cell line MCF-7 SP subsets are different content, to provide scientific reference value for the identification of cancer stem cells and purification. 2 contains a higher proportion of SP cells in the human breast cancer cell line MCF-7, and the SP and the NSP cells were identified and purified antigen differences.
