Abstract


Objective To establish suis infected BALB / c mouse model, detection of BALB / c mice peripheral blood and spleen cytokine interleukin -17 (IL-17), β transforming growth factor (TGF-beta), leukocyte dynamic changes in interleukin -6 (IL-6), to explore the role of Th17 subsets in disease occurrence, development and prognosis infection caused by the body's inflammatory response and immune response mechanism to provide a theoretical basis for proven Eperythrozoon of . Liaoning Medical College of Animal Husbandry and Veterinary pig blood collected aseptically 20 were identified by microscopy and PCR the Eperythrozoon positive blood, spare. BALB / c mice were randomly divided into group A (purified the Eperythrozoon group), B group (positive experimental group), C group (negative experimental group), D (control group), four groups of mice were intraperitoneally were inoculated 0.5 ml of purified Eperythrozoon suis-positive blood samples suis-negative blood samples, normal saline. After inoculation 1d, 3d, 5d, 7d, 9d, ELISA was used to detect the peripheral blood of mice, IL-17, TGF-β, IL-6 level changes detected by RT-PCR of mouse spleen IL-17, TGF -β, IL-6 mRNA expression. Results 1 ELISA and RT-PCR detection of IL-17 in the results shown in the first day after vaccination, 3d, 5d, 7d, 9d, A group higher than the D group (P lt; 0.05), B group than in the C group, group D (P lt; 0.05), C group than in group D (P lt; 0.05), and reached a peak in the first 5d of vaccination. 2, ELISA, and RT-PCR to detect the level of TGF-β results shown after inoculation, 1d, 3d, 5d, 7d, A group than in group D (P lt; 0.05) in group B than group C, D group ( P lt; 0.05), C group than in group D (P lt; 0.05), and peaked inoculation 3d, 9d resume normal. 3, ELISA and RT-PCR to detect the level of IL-6 results shown in the first day after vaccination, 3d, 5d, 7d, 9d, A group than in group D (P lt; 0.05) in group B than in group C, D group (P lt; 0.05), C group than in group D (P lt; 0.05), and reached a peak in the vaccination section 7d. 4, correlation analysis, ELISA Methods: IL-17, IL-6 of both trends positive correlation of the group R A = 0.420, B group of r = 0.456, TGF-beta, IL-6 both trends were negative correlation, the group A r = -0.431, B group R = -0.576; RT-PCR method: IL-17, IL-6 both trends positive correlation group R A = 0.568, B-group R = 0.639 and TGF -β, IL-6 both trend was negatively correlated with the group A r = -0.645, B group R = -0.829. Conclusion 1. Suis infected BALB / c mice, an animal model experiments successfully established and dynamic observations of each time period BALB / c mice peripheral blood and spleen of IL-17, TGF-β, IL-6 changes in the level. 2. BALB / c mice were infected suis can occur early IL-17, TGF-β, and IL-6 levels phenomenon. The prompted the Eperythrozoon causing organism Th17 cell subsets of the differentiation, the findings provide scientific data for proinflammatory mechanisms eperythrozoonosis. 3, IL-17, TGF-β, IL-6 expression levels of the peak, respectively 5d, 3d 7d, results suggest that the relationship between the three, i.e., TGF-β, and IL-6, under the joint action to promote Th17 secrete IL -17, IL-17 generation can contribute to other cells to secrete IL-6. This is the theory of dynamic data Th17 subsets in the process of Eperythrozoon infection.
