Background:Hypertrophic scar was pathological scar mainly caused by trauma which injured to dermis characterized by the excessive proliferation of fibroblasts and extracellular matrix excessive production and deposition. Hypertrophic scar could cause significant defects in appearance or dysfunction depended on different growth position. Hypertrophic scar was the major problem which trauma patients needed to face during post-rehabilitation. The prevention and cure of hypertrophic scar was always a difficult matter but we were not clear on the mechanism of hypertrophic scar so far. PPAR-βwhich belonged to PPARs family was rich in skin. The research showed that PPAR-βwas important to the process of wound healing. Hypertrophic scar was considered to be the result of excessive wound healing, but the studies on PPAR-βin the formation and development of hypertrophic scars was few.Objective:To compare the different expression of PPAR-βbetween human hypertrophic scar and normal skin tissue. To provide a new research direction on the formation and development of hypertrophic scar and a new strategy of curing hypertrophic scar. Methods:The specimen of hypertrophic scar and the corresponding normal skin tissue was obtained from 16 patients underwent scar surgery from January 2010 to January 2011 in Burns and Plastic Surgery of Xiangya hospital of Central South University. RT-PCR was performed for semi-quantitative analysis of the expression of PPAR-βmRNA in hypertrophic scar and normal skin. Immunohistochemistry was performed to determine the expression and localization of PPAR-βprotein in the hypertrophic scar tissues and to further verify the different expression of PPAR-βin tissueResults:RT-PCR analysis result indicated that PPAR-βmRNA was expressed in hypertrophic scar and normal skin tissue but the expression level of PPAR-βmRNA was higher in hypertrophic scar tissues than that in normal skin tissue with significant difference (t=2.361,P=0.028). The result of immunohistechermical analysis indicated PPAR-βprotein mainly located in the cytoplasm of fibroblasts cells and there were 12 patients with PPAR-βpositive expression in hypertrophic scar specimens (positive rate 75%) compared that 5 patients in normal skin tissue specimens (positive rate 31.2%). the expression level of PPAR-βprotein was higher in hypertrophic scar tissue than that in normal skin tissue with significant difference (X2=6.14 P=0.013)Conclusion:We firstly demonstrated the higher expression of PPAR-βin hypertrophic scar tissues than normal skin tissues with significant difference. Our results indicated that PPAR-βmay play an important role in the formation and development of hypertrophic scar
