The Effect of Sustained Hyperuricemia on Endothelial Function in Rats

Abstract


ObjectiveTo investigate sustained hyperuricemia effect of the function of endothelial cells, and further research on mechanism of the dysfunction of endothelial cells.MethodsHyperuricemia models were induced in 20 male Sprague Dawley rats for 12 days with uricase inhibitor, Oxonic acid (750 mg/kg/day), by gavage. Blood was collected by puncturing of the retrobulbar venous plexus at 0,4th,8th and 12th day, and sent to test serum uric acid and creatinine by the methods of Trinder’s and Jaffe, respectively, through the hitachi automatic 7180 biochemical analyzer determine. Endothelial nitric oxide synthetase (eNOS) and nitric oxide (NO) was measured by the points photometry instrument. Rats were sacrificed at 12th day after blood collection. Urate deposition was investigated in abdominal aorta and kidney with HE staining under electronic light microscopy. Measurements were analyzed by paired-samples T test.ResultsWhen blood was collected by puncturing of the retrobulbar venous plexus, there are six rats died in respiratory inhibition, and four rats died in eye infections. Oxonic acid induced hyperuricemia at 4th day (p=0.004); the mean value of uric acid was lower at 8th day compared with 4th day, but still higher than base line; uric acid increasing again at 12th day and significantly higher than base line (p=0.014). The level of nitric oxide and endothelial nitric oxide synthetase decreased steadily, respectively. Compared with base line, the level of nitric oxide was not significantly at 4th day and 8th day, there was significant difference between base line and 12th day (p=0.009). The level of endothelial nitric oxide synthetase was difference at 8th day (p=0.012), and significant difference at 12th day (p=0.004).The level of serum creatinine was not significant difference at 0,4th,8th and 12th day. Urate deposition was not observed in the abdominal aorta and kidney samples.ConclusionThe uric acid increases are not associated with renal insufficiency, the levels of nitric oxide and endothelial nitric oxide synthetase decreased is not coupling. Hyperuricemia induced endothelial dysfunction in time-dependent manner, which may result in the cardiovascular diseases.
