Freshwater bacterial sepsis (Systemic Bacterial Septicemia) is the history of our country harm aquaculture fish species most, the most widely popular waters, causing serious economic losses of an acute infectious disease, has restricted the healthy development of freshwater aquaculture in One of the most important diseases. Antibiotics and other chemicals commonly used in the presence of many drawbacks, makes finding a safe and effective alternatives to antibiotics become an urgent problem. In this study, bacterial sepsis caused major fish pathogens - Aeromonas hydrophila (Aeromolla hydrophila, A. hydrophila) as antigen-specific immune Lohmann hens get IgY; IgY separation, purification, measured purity and specific antibody titer levels and in vitro antibacterial, immunofluorescence experiments in vitro on specific IgY Aeromonas hydrophila growth inhibition was studied; through oral, immersion and injection, investigate specific IgY Heilongjiang suffering from bacterial sepsis treatment and prevention of silver carp effect. The results show that, diluted with water - salting - gel filtration method of extraction and purification of specific IgY purity of 89.3%, water-soluble components (1:6 dilution) total IgY content of 9.47 mg / mL, specificity immune IgY content with the extension of time was significantly higher, up to 1.25mg/mL, total IgY content of 13.19%; specific IgY titers of up to 1:12800. Immunofluorescence showed that specific IgY with Aeromonas hydrophila strong binding effect. Experimental results show that in vitro antibacterial 75mg/ml specific IgY, could significantly inhibit Aeromonas hydrophila in liquid medium growth (P lt; 0.05). The specific IgY with pathogens crude extract mixed at a 1:1 ratio, intraperitoneal injection of healthy Heilongjiang silver carp experimental results show that compared with the control group of specific IgY infected fish can reduce the cumulative mortality rate around 40%. Investigate specific IgY fed diets containing different proportions of feed before and after the exchange that poison Heilongjiang silver carp protective and therapeutic effects visible, add 5% passive protective effect of specific IgY best, the cumulative mortality rate of about 50%; 0.5% specificity the therapeutic effect of IgY worst, the cumulative mortality rate was 80%; investigated with different concentrations of silver carp Heilongjiang IgY solution soak after pathogen attack drug use and the protective effect after challenge with different concentrations of IgY solution soak the therapeutic effect of fish Visible, soaked in 400mg/mL specific IgY solution best prevention group, the mortality rate was 30%; investigated after injection of different doses of IgY pathogen attack drug use and the protective effect after challenge injection of different doses of the therapeutic effect of IgY display , injection 4mg / tail specific IgY better prevention, mortality was 50%. At the same dose levels, regardless of the mode of administration, are superior therapeutic effect preventive effect. The results show that specific IgY preparation of anti A.hydrophila sepsis Heilongjiang silver carp have a preventive effect.
