TRMP7 mRNA expression in gastric carcinoma and clinical significance Preface gastrointestinal malignancies is one of the common malignant tumors in China, one of the most common gastrointestinal cancer often leads to poor prognosis, seriously endangering the life and health of the patient. Transient receptor potential (transient recepter potential, TRP) is a class of non-voltage-dependent cation channel widely expressed in a variety of biological tissue and cells, and to participate in a variety of important physiological functions, including muscle contraction, neurotransmitter release, cell proliferation, cell differentiation, gene transcription, apoptosis and angiogenesis. TRPM7 as a widely expressed in the permeability of the channel protein TRPM subfamily member of the TRP family of Ca2 and Mg2. The research reports of TRPM7 cation channel cell Mg2 balance, adjustment of bowel movements, cell survival and growth, synaptic transmission and cellular adhesion to a variety of physiological functions. In recent years, of TRPM7 channel relationship between the tumor has become the focus of attention of the researchers, the study found TRPM7 channels play an important role in the survival, growth and proliferation of human mast cells in head and neck cancer cells and osteoblast-like cells. Recently, Kim et al found that the TRPM7 channel of human gastric adenocarcinoma cell survival is critical. At present, domestic and foreign TRPM7 expression in gastric cancer and correlation with biological behavior of gastric cancer reported in the literature are very few, so this study by detecting gastric TRPM7 mRNA expression levels of the corresponding adjacent normal tissue, to explore TRPM7 and gastric pathology biological behavior. Materials and methods experimental material 1,48 cases stored in liquid nitrogen fresh gastric cancer tissues and corresponding adjacent normal gastric tissue (confirmed by pathology without cancer invasion). 2, tissue total RNA extraction and RT-PCR reagents. 3, Real-time PCR reagents. Experimental methods, according to the of TRPM7 and β-actin gene sequence design primers. 2, total RNA was extracted 48 cases of gastric cancer and adjacent stomach tissue and reverse transcription and PCR amplification. 3, Real-time PCR detection TRPM7 mRNA expression in gastric cancer and adjacent normal gastric tissue. 4, statistical analysis of the relationship of TRPM7 with gastric cancer pathobiological behavior. Results 1, TRPM7 mRNA are expressed in normal gastric tissue and gastric cancer tissue. 48 patients with gastric cancer, 38 cases TRPM7 gene mRNA expression was down 77.1% (37/48); 11 cases TRPM7 gene mRNA is increased, accounting for 22.9% (11/48). Gastric carcinoma and adjacent normal tissue TRPM7mRNA the relative expression levels (ACT value) is 5.12 ± 0.18 and 4.32 ± 0.19, respectively, compared carcinoma and adjacent normal gastric tissue, TRPM7 gene mRNA expression was significantly reduced in gastric carcinoma. 2, in the lymph node metastasis of gastric carcinoma, TRPM7 gene expression than without metastasis cancer tissue significantly enhanced. Conclusions of gastric carcinoma TRPM7 the genes downregulated prompt its participation in the pathogenesis of gastric cancer and may be associated with lymph node metastasis occurred related.
