Abstract


Background: Colorectal cancer is a common gastrointestinal malignancy, has the characteristics of high recurrence and metastasis. The 5-year overall survival rate of about 60% in recent years, the incidence gradually increased, seriously endangering the health of the people's living. The incidence of colorectal cancer and its to transfer no effective means of prevention and treatment, pathogenesis and prevention research in this area is still hot. Anx A1 is one of the members of the calcium phospholipid binding protein Anx superfamily in signal transduction, DNA replication, cell cycle regulation, cell transformation, and play an important role in many physiological processes of apoptosis, and the formation of the calcium channel. Many studies have confirmed AnxA1 with the occurrence of a variety of tumors are closely related, and its expression in multiple tumor significantly abnormal, directly or indirectly involved in tumorigenesis. Purpose: The purpose of this study is to apply immunohistochemistry, fluorescence quantitative RT-PCR and Westernblot detected peritumoral normal mucosa, colorectal precancerous lesions (tubular villous adenoma) of Anx A1 in the different levels of differentiation of colorectal cancer and colorectal cancer lymph node metastasis foci expression levels, in order to explore the relationship between Anx A1 expression levels and the incidence of colorectal cancer progression and metastasis. Methods: We collected 20 cases of colorectal cancer and 10 cases of tubular villous adenoma fresh specimens, using immunohistochemical SP method, fluorescence quantitative RT-PCR and western blot method to detect the expression of of Anx A1 in mRNA and protein levels. Further collection of paraffin-embedded tissue blocks of colorectal cancer, 30 cases by immunohistochemistry SP method detection Anx A1 in normal colorectal mucosa, tubular villous adenoma, the degree of differentiation of colorectal cancer as well as the same sample of the primary tumor and lymph node metastatic foci in the expression level of the Anx A1. SPSS statistical software for data processing and statistical analysis. The results: a, the immunohistochemistry and Westernblot detect AnxA1 protein in normal colorectal mucosa, tubular villous adenoma, colorectal cancer and its lymph node metastasis foci in the expression and clinical significance AnxA1 in intestinal epithelial cells mainly located in the cell extracellular pulp. Adenoma, adenocarcinoma and adjacent normal tissues have different levels of positive expression, yellowish, brownish-yellow or brown particles. Adenocarcinoma, adenoma and normal mucosa of group AnxA1 expression differences were significant (H = 16.913, P = 0.000 lt; 0.05), overall reflect this trend reduce AnxA1 expression intensity increased with the degree of tumor differentiation. Lymph node metastasis group expression of the AnxA1 in lymph node metastases is stronger than the primary tumor, with a significant difference (H = 4.79,0.025 lt; P lt; 0.05), and Anx A1 expression and lymph node metastasis were positively correlated (Z = 0.5702 ). Second, fluorescence quantitative RT-PCR was used to detect the of Anx A1m RNA in normal colorectal mucosa, tubular villous adenoma, colorectal cancer and its lymph node metastasis of carcinoma and clinical significance at the mRNA level, Anx A1 in the intestinal mucosal epithelial widely expressed, colorectal cancer group expression compared with normal mucosa down, three groups express differences significant resistance (H = 24.338, P = 0.000 lt; 0.05), of Anx A1 mRNA in expression and large bowel cancer in different differentiation degree of relevant (H = 9.568, P lt; 0.05 ), but its expression in colorectal cancer lymph node metastasis (Z = -0.908) correlation. No significant differential expression of lymph node metastasis and without lymph node metastasis in colorectal significance (P = 0.155 gt; 0.05). Conclusion 1 of Anx A1 protein low expression in adenomas and adenocarcinomas in the normal mucosa upregulated expression levels with the degree of tumor differentiation; and its correlation with lymph node metastasis were positively correlated; 2.Anx A1 mRNA in intestinal mucosa widely downregulated expression in adenocarcinoma, and mRNA expression levels change before the tumor morphological changes, suggesting that the change tumor may be relevant; 3.Anx A1 expression and colorectal cancer development and metastasis, AnxA1 The protein may be one important indication as to reflect the biological behavior and prognosis of colorectal cancer.
