Background systemic lupus erythematosus (SLE) is a complex clinical manifestations, can affect multiple organs and systems of the kidneys, lungs, heart, blood, serum appear in a variety of autoantibodies in autoimmune diseases. SLE pathogenesis is still not yet clear, the occurrence of this disease with genetic, environmental, immunological, viral infection, drugs and hormones, and other factors, is generally believed that individuals with a genetic predisposition factors of the environment, sex hormones, and infection caused immune function abnormal autoantibody production, immune complex formation and deposition in the organization, leading to the occurrence and development of systemic lupus erythematosus. Genetic factors play an important role in the pathogenesis of SLE. SLE susceptibility genes discovered through genome-wide scan and candidate gene approach. Graphics program (Human Haplotype Map, HapMap) as the international monomer to complete efficient and inexpensive high-throughput genotyping technologies emerge in recent years, genome-wide association analysis (Genome-wide association study, GWAS) of different races The population of SLE patients found that the number of new susceptibility genes and loci. It is noteworthy that in the Han population in Europe and China, respectively, reported two TNIP1 gene single nucleotide polymorphisms (Single nucleotide polymorphism, SNP) rs7708392 and rs10036748 with SLE susceptibility was significantly related (P = 4.5 × 10 - 7; P = 1.67 × 10-9). As tumor necrosis factor-α-induced protein 3 (TNFAIP3) interacting protein, TNIP1 the collaborative TNFAIP3 gene inhibits NF-κB activation and TNF-mediated apoptosis. In this study, the use of real-time quantitative PCR technique to explore the role of TNIP1 gene in the pathogenesis of SLE analysis TNIP1 gene mRNA expression in cases and control. Objective To investigate TNIP1 gene mRNA expression in peripheral blood mononuclear cells (PBMCs) of SLE patients, as well as the correlation with SLE disease activity index (SLEDAI) and susceptibility loci rs10036748. Collected 66 cases of 61 cases of patients with SLE and control, mRNA was extracted from peripheral blood mononuclear cells (PBMC) and reverse transcribed into cDNA, adding the corresponding Taqman primers and probes using the ABI Prism 7900 high-throughput quantitative PCR The detect the PBMC TNIP1 mRNA expression the situation; application the Sequenom MassArray technology in all cases control the SNP rs10036748 point genotyping; SPSS13.0 software for statistical analysis of data. Results SLE patients TNIP1 gene mRNA expression is lower than the normal control group, the difference was significant (P lt; 0.05); TNIP1 gene mRNA expression levels with SLEDAI and SNP rs10036748 related to sex (P gt; 0.05). Conclusion TNIP1 gene expression abnormalities suggesting that this gene may play an important regulatory role in the pathogenesis of SLE process.
