Abstract


Objective: To study the human early pregnancy villous and decidual matrix metalloproteinase -9 (MMP-9) and tissue inhibitor of matrix metalloproteinase -1 (TIMP-1), the positioning and expression of different doses of mifepristone is detected its expression impact; explore the role of MMP-9 and TIMP-1 in the mifepristone early pregnancy mechanism, and the relationship of both normal early pregnancy. Methods: normal early pregnant women undergoing abortion study 60 cases were randomly divided into four groups: control group, the 50 mg group, 100mg, 150mg, each group of 15 cases. Control group without medication direct line to the vacuum aspiration of the uterus, the experimental group mifepristone 50mg, 100mg, 150mg 48 hours underwent vacuum aspiration get chorion and decidua using immunohistochemical techniques and computer image analysis system to detect MMP-9 and TIMP-1 localization and expression. Results: 1, MMP-9 and TIMP-1 in the four groups of the villous trophoblast cells, the cytoplasm of capillary endothelial cells, decidual decidual cells, the cytoplasm of glandular epithelial cells and vascular endothelial cells showed varying degrees of positive staining. 2, four groups of villi and decidua MMP-9 and TIMP-1 expression 2.1 the chorionic villi medication groups compared with the control group, the expression of MMP-9 was significantly enhanced (P lt; 0.01), TIMP-1 expression was significantly weakened (P lt; 0.01) of MMP-9/TIMP-1 ratio was significantly higher (P lt; 0.01); medication among groups of MMP-9 and TIMP-1 expression was no significant sex difference (P gt; 0.05 ), MMP-9/TIMP-1 and medication dose was positively correlated (P lt; 0.05). 2.2 decidua medication groups compared with the control group, the expression of MMP-9 significantly increased (P lt; 0.01), TIMP-1 expression was significantly reduced (P lt; 0.01) of MMP-9/TIMP-1 ratio significantly higher (P lt; 0.01); expression of MMP-9 in the 150mg group than in the 50mg group and the 100 mg group was significantly enhanced (P lt; 0.01), 50mg and 100mg group, no significant difference (P gt; 0.05); expression of TIMP -1 expression and medication dose was negatively correlated (P lt; 0.01); MMP-9/TIMP-1 in medication dose was positively correlated (P lt; 0.01, P lt; 0.05). Conclusion: Mifepristone can affect the villi and decidual MMP-9 and TIMP-1 expression, the expression of MMP-9 enhanced TIMP-1 expression was decreased to excessive degradation of extracellular matrix fluff peel, slough membrane tissue stripping. 2, MMP-9/TIMP-1 villi and decidual medication in each group, the ratio was significantly higher than in normal pregnancy, and the ratio was positively correlated with the dose, MMP-9 and TIMP-1 balance for normal early pregnancy Maintain play an important role.
