Objective: Gastric cancer is the most common malignancy in the country of its high mortality rate 25.2/10 Wan. The occurrence and development of gastric cancer is a multi-gene, multi-step dynamic evolution process that involves the activation of multiple oncogenes, tumor suppressor gene inactivation, and the resulting cell biology changes in behavior. Clinical studies show that approximately 60% of patients with gastric cancer was going through aggressive surgery and (or) drug treatment, but eventually died of recurrence and metastasis. Therefore, studies of gastric cancer, and to explore the molecular mechanism of the development of new therapeutic measures is extremely important. Evolution of the tumor is a tumor cell with the surrounding extracellular matrix (extracellular matrix, ECM) interactions between the disorder is characterized by severe. Membrane-type matrix metalloproteinase 1 (membrane type matrix metalloproteinase-1, MT1-MMP) can degrade many ECM components, resulting in the release of a variety of biologically active molecules, and by controlling cell adhesion and cytoskeleton regulate cell migration build , growth, differentiation and survival. Studies have shown that, MT1-MMP expression levels are closely related with the development of tumors, tumor biological characteristics that affect one important factor. Kazal motif cysteine ??rich protein induced reverse (reversion-inducing-cysteine-rich protein with kazal motifs, RECK) Takahashi et gene v-Ki-ras-transfected NIH3T3 cells by expression cloning method of separation of a transcriptional suppressor gene. RECK through a variety of mechanisms inhibiting the secretion and activity of a variety of MMPs, thereby inhibiting tumor invasion and metastasis. In addition, RECK for the developing process of angiogenesis is essential, it will result in the reduction of tumor angiogenesis and thus promote tumor evolution. In this study, patients with gastric cancer as the research object, using RT-PCR method for detection of MT1-MMP and RECK mRNA in gastric carcinoma and normal gastric mucosa, analyze these indicators and biological behavior of gastric cancer to investigate the occurrence of gastric cancer, invasion and metastasis development role in the process. Methods: Fourth Hospital of Hebei Medical University, outside the three subjects in September 2006 - June 2007 surgical specimens of gastric cancer were 40 cases. Were collected and normal gastric mucosa (more than 5cm from the edge of the tumor), postoperative were confirmed by pathology. All patients had not received chemotherapy or biological therapy and other interventions. Reverse transcription - polymerase chain reaction (reverse transcription polymerase chain reaction, RT-PCR) technology to detect gastric cancer and normal gastric mucosa MT1-MMP and RECK mRNA expression, expression analysis of two patients with gastric gender, age, tumor location, tumor size, gross type, degree of differentiation, depth of invasion, lymph node metastasis, clinical stage and other clinical biological behavior, and?-actin as an internal reference standard. All data applications SPSS12.0 statistical software for statistical analysis, P lt; 0.05 statistically significant. Results: 1 gastric MT1-MMP mRNA expression levels (0.90 ± 0.03) was significantly higher than that in normal gastric mucosa (0.76 ± 0.10), the expression level (P lt; 0.05). 2 gastric RECK mRNA expression levels (0.81 ± 0.04) was significantly lower than that in normal gastric mucosa (1.01 ± 0.14), the expression level (P lt; 0.05). 3 MT1-MMP mRNA gastric cancer invasion T3 T4 group in the expression levels (1.04 ± 0.07) than T1 T2 group expression levels (0.60 ± 0.06), the difference was statistically significant (P lt; 0.05); in with lymph node metastasis expression levels (1.10 ± 0.23) than those without lymph node metastasis group expression levels (0.59 ± 0.01), the difference was statistically significant (P lt; 0.05); in the clinical staging of group III-IV expression levels (1.06 ± 0.14) than stage I-II expression level of the group (0.67 ± 0.10), the difference was statistically significant (P lt; 0.05). 4 RECK mRNA in gastric cancer invasion T3 T4 group, the expression levels (0.60 ± 0.09) than T1 T2 group, the expression levels (1.04 ± 0.17), the difference was statistically significant (P lt; 0.05); in lymph node The expression levels of metastasis group (0.56 ± 0.06) than those without lymph node metastasis group expression levels (1.00 ± 0.11), the difference was statistically significant (P lt; 0.05); in the clinical staging of group III-IV expression levels (0.56 ± 0.08) than stage I-II expression level of the group (1.03 ± 0.03), the difference was statistically significant (P lt; 0.05). 5 gastric cancer MT1-MMP and RECK mRNA levels and patient gender, age, tumor location, tumor size, gross type and histological grade. 6 gastric cancer tissues of MT1-MMP and RECK mRNA levels have a negative correlation (rs = -0.29), but not statistically significant. Conclusion: a gastric carcinoma MT1-MMP mRNA expression levels and RECK mRNA expression levels decreased, indicating that MT1-MMP involved in gastric carcinogenesis, RECK expression reduces to promote the development of gastric cancer. MT1-MMP and RECK mRNA can be used as research development of gastric cancer clinical and biological indicators. 2 gastric cancer MT1-MMP mRNA expression levels with the deepening of the depth of invasion, lymph node metastasis, clinical stage increases significantly increased (p lt; 0.05). Description increased expression of MT1-MMP in gastric cancer invasion and metastasis and clinical pathology related to the process. 3 gastric cancer RECK mRNA expression levels with the deepening of the depth of invasion, lymph node metastasis, clinical stage and significantly lower (p lt; 0.05). Description RECK expression reduces invasion and metastasis of gastric cancer and clinical pathology related to the process. 4 gastric cancer MT1-MMP and RECK mRNA levels and patient gender, age, tumor location, tumor size, gross type and histological grade. 5 gastric cancer between MT1-MMP and RECK a certain antagonism. Additional cases pending further research lines.
