Abstract


Objective To investigate the survivin (survivin) in neuroendocrine technology (NED) prostate cancer (PCa) Expression and significance analysis survivn expression of neuroendocrine prostate cancer (NED) and its correlation with the clinical stage of prostate cancer, the relationship between histological grade, as well as resistance to endocrine therapy for the treatment of prostate cancer, the prospects look to. Methods Third Xiangya Hospital, Central South University, Department of Pathology, January 2003 - November 2007 between the diagnosis of prostate cancer and benign prostatic hyperplasia (BPH) cases and its clinical and pathological data, complex reading HE staining histopathology. To observe the three proteins in the same tissue, the present experiment using a continuous tissue sections were observed in different parts of the same organization the same protein expression. Prostate cancer mainly depend on the degree of neuroendocrine secretory product of its detection, the present study, immunohistochemical staining of peroxidase-labeled streptavidin method (SP method) combined detection of synaptophysin (SYN) and chromaffin granules Protein A (Cga) expression, in order to improve neuroendocrine oriented (NED) detection of the extent to which the SYN and / or diagnosis of CgA positive expression of neuroendocrine prostate cancer. While using immunohistochemical SP method survivin expression in this group was observed survivin in neuroendocrine carcinoma of the prostate condition. Results (1) 28 cases in PCa, SYN, CgA and survivin positive expression rates were 53.6%, 50.0%, 89.3%, SYN and CgA expression in BPH were negative, survivin in benign prostatic hyperplasia (BPH) expressed rate of 11.1%. Three proteins were positive expression in PCa group with positive expression in the BPH group, the difference was significant (P <0.05). (2) 28 cases of PCa in 25 cases (89.3%) survivin expression, 21 cases (75%) SYN and / or CgA expression, survivin in 21 cases of SYN and / or CgA positive PCa (ie of prostate neuroendocrine cancer) in 20 cases (95.2%) were positive, survivin positive expression NED with prostate cancer was positively correlated (correlation coefficient r_s> 0). (3) in 28 cases of PCa in 14 cases (50%) CgA expression was positive in 15 cases (53.6%) SYN expression was positive, both of which eight cases were positive; while CgA and / or SYN positive in 21 cases (75%), showing that combined detection of Chromogranin A (CgA) and synaptophysin (SYN) significantly improved neuroendocrine technology (NED) detection rate. (4) PCa group survivin expression with age regardless of> 70 years and <70 years were compared no significant difference (P> 0.05); while with clinical staging and grading, low-differentiated group and high differentiated group, the difference was significant (P <0.05). Conclusions (1) SYN and Cga combined detection of prostate cancer can significantly improve the detection of neuroendocrine degree. (2) survivin expression in prostate cancer tissue grading and TNM stage, namely the higher grade, higher expression; clinical stage later, the expression is higher. (3) survivin and neuroendocrine PCa was positively correlated with the degree, the higher the expression of neuroendocrine higher the degree; due to neuroendocrine survivin inhibits apoptosis, therefore prostate cancer may lead to increased expression of survivin neuroendocrine technology; Neuroendocrine cells produce cytokines and the role of neuropeptides neuroendocrine cells and the surrounding non-tumor cells, resulting in the growth of androgen-independent, and thus resistance to endocrine therapy for prostate cancer.
