Abstract


Objective(1)To establish a method to extract,purify and test steroid hormone from rat brain;(2)To determine the level of estradiol in serum and hippocampus in different estrous cycle of female rat;(3)To dynamically observe the level of estradiol in serum and hippocampus after the onset of epilepsy and the relationship between the level of estradiol and severity of epilepsyMethods(1) The rat were divided into five groups:proestrus group,estrus group,metestrus group,diestrus group of female rat and male group;each group has six rat and serum and hippcampus samples were collected;(2) The seizure model was established by kindling of amygdale on estrus female rat; latency of seizure,time to severe stage,the fequency and duration of seizure were recorded;Serum and hippocampus samples 1h,2h,3h,4h,5h,6h,12h,24h after seizure were collected respectively;(3) Method for steroid hormone detection:①homogenization of hippocampus tissus;②extraction of steroid hormone by organic solvent;③purification of steroid hormone by HPLC;④detection of steroid hormone by RIA or ELISA.Results(1)The highest level of estradiol in the serum and hippocampus were found in the proestrus group and estrus group respectively,there were statistically significant differences with the other groups.Statistically significant differences were found between the male group and the female group.(2)The concentration of estradiol in the hippocampus was much higher than that in the serum which was approximately two to seven times greater than that in the serum,with the hightest hippocampus/serum ratio in the estrus group and the lowest hippocampus/serum ratio in the proestrus group.(3)Statistically significant differences of hippocampus level of estradiol were found between the KA group and the NS group of the same time point after seizure onset,while no correspending difference were found in the serum.(4)Approximately 1～2 minutes after KA intra-amgydale injection the rat began to revival from anesthetization,5～10 minutes later the rat began to show epileptic paroxysm.The peak were reached in 180 minute after KA injection and then the epileptic paroxysm was alleviated.The correlation of seizure severity scale to hippocampus level of estradiol was found.(5)No statistically significant differences of hippocampus level of pregnenolone were found between the KA group and the NS group of the same time point after seizure onset.Conclusions(1) Hippocampal levels of estradiol are a result of peripheral production as well as synthesis within the brain,and the main source is de novo synthesis in the hippocampus.(2) There were dynamic changes of hippocampal level of estradiol after seizure onset which had consistency with seizure severity scale and no statistically significant change of serum level of estradiol were found which indicated the elevation of hippocampus level were mainly synthesis de novo.(3) No correlation of hippocampus level of pregnenolone and estrous cycle or seizure onset was found which indicated hippocampus estradiol may not synthesis from pregnenoloen but from androgen by aromatase
