Abstract

Purpose by the gap junction protein (connexin, Cx) constitute the cell gap junction (gap junction) play an important role in the regulation of cell proliferation and differentiation, Cx protein expression down to cause the loss of function of the cell junctional intercellular communication (gap junction intercellular communication, GJIC) of cell metabolism, proliferation, differentiation and apoptosis of the loss of cells in the uncontrolled proliferation and differentiation state of disorder and eventually deteriorate. Epithelial cadherin (E-cadherin), inhibit its function or expression decreased cell adhesion capacity declined allows easy cell separation shedding of epithelial cells and between cells and cell-extracellular matrix adhesion transmembrane glycoprotein molecules , followed by the formation of invasion and metastasis. The purpose of this study is to investigate the connexin 43 (Connexin 43, Cx43) and E-cadherin expression in human colorectal cancer and their mutual relations. Methods SP immunohistochemical method in 60 cases of colorectal cancer tissues Cx43 and E-cadherin protein expression, analysis of the clinical significance of the expression and correlation analysis between the two. The results showed a typical membrane expression of Cx43 and E-cadherin protein in 16 cases of normal colorectal tissues, the positive rate was 100% good functional gap junctional intercellular cell adhesion exists between prompts cells. 60 cases of cancer tissue, only 33 cases of the Cx43 positive expression (55%), 24 cases of expression of E-cadherin-positive (40%), indicating that the Cx43 and E-cadherin protein expression in normal tissue and cancer tissue were significantly differences and positive expression of Cx43 and E-cadherin significantly weakened mainly cytoplasmic expression. Cx43 and E-cadherin positive expression rate of cell differentiation, Dukes stage and lymph node metastasis in 60 cases of cancer tissue are closely related, has nothing to do with the histological type, gender and age. With the reduction of the degree of differentiation, Cx43 and E-cadherin positive expression rate also decreased significantly (P lt; 0.01, P lt; 0.05); different Dukes stages Cx43 and E-cadherin positive expression rate with the installments increased and decreased (P lt; 0.01, P lt; 0.01); Cx43 and E-cadherin positive expression rate of lymph node metastasis in cancer tissue than those without lymph node metastasis of cancerous tissue significantly reduce (P lt; 0.01, P lt; 0.01). 21 cases out of 60 cases of cancer of Cx43 and E-cadherin positive expression, 24 patients with negative expression, E-cadherin positive expression of Cx43 negative expression of three cases of positive expression of Cx43 and E-cadherin-negative expression of the 12 cases, statistics The analysis showed significant differences (rs = 0.53), indicating that Cx43 and E-cadherin protein expression has significant correlation. Conclusion Cx43 and E-cadherin protein expression in colorectal dropped significantly, and there is a clear correlation between both, may be a common event in colorectal cancer occurrence and development process.
