Growing synthetic substances of environmental pollution and toxicity problems, people are increasingly concerned about the needs of plant natural products. The active ingredient in the plant has become the major research object in one of the people. Flavonoids (flavonoids) are a class exists in nature, with 2 - phenyl chromone (flavone) the structure of the compound, and an important source for the development of natural antioxidants. This article in the Jerusalem artichoke leaves as raw material, through single factor and orthogonal experiments designed to explore the optimum extraction conditions of the Jerusalem artichoke leaves flavonoids. And Jerusalem artichoke leaf flavonoids extract biological activity, including stability, oxidation resistance, anti-tumor activity and antibacterial activity of a preliminary study. The main content and the results are as follows: 1. Jerusalem artichoke leaf flavonoids extraction process using three methods of jerusalem artichoke leaves flavonoids extracted, the best extraction process were: of ethanol reflux method extraction Jerusalem artichoke leaf flavone best conditions: 80% ethanol concentration, solid-liquid ratio of 1:35, extraction temperature of 90 ° C, extraction time of 2 h, this condition flavonoids was 1.411%. Jerusalem artichoke leaves flavonoids ultrasound-assisted ethanol extraction conditions: 60% of the solid-liquid ratio 1:20, ethanol concentration, extraction time of 30 min, extraction temperature of 80 ° C, 1.921% of the extraction rate of total flavonoids in this condition. Jerusalem artichoke leaf flavone microwave-assisted ethanol extraction conditions: 50% ethanol concentration, extraction time of 5min, solid-liquid ratio 1:30, microwave power 300W, this condition flavonoids was 2.481%. Compare three different extraction methods, microwave extraction of flavonoids highest rate, the lowest concentration of ethanol greatly shorten the extraction time, so microwave extraction, microwave extraction are used later in the trial. Jerusalem artichoke leaf flavonoids biological activity of stability experiments, Jerusalem artichoke leaf flavonoids have a certain amount of heat resistance, but should avoid high temperature, strong base and redox should save the best sealed dark at room temperature. The antioxidant experiments show that the Jerusalem artichoke leaf flavonoid extract of superoxide anion, hydroxyl radical and DPPH radicals have certain clear role and clear the effect of the extract to the increase of the amount of the increase. Jerusalem artichoke leaf flavonoids effect against lipid peroxidation. The anti-tumor effect of extract concentration 200μg/mL human nasopharyngeal carcinoma cells (KB), human hepatoma cells (HepG-2) in human leukemia cells (K562) showed some inhibition. The combination of inhibition zone experiment results comprehensive view of the minimum inhibitory concentration (MIC), inhibition of largest to smallest as follows: Staphylococcus aureus gt; E. coli gt; Bacillus subtilis.
