Abstract

Purpose: The application of different experimental methods to detect immunoglobulin heavy chain binding protein (Bip) 2 cells expression in human osteosarcoma cell line Saos , and further study in 20 patients with osteosarcoma clinical specimens expression; surrounding normal tissue and compare the differences in the expression of osteosarcoma tissue and normal tissue; preoperative chemotherapy, preoperative chemotherapy specimens express the difference; distant metastasis and the expression differences between the specimen is not transferred. And so to infer this Bip expression in osteosarcoma osteosarcoma growth and metastasis of the relationship as well as the effects of chemotherapy on the expression, in order to provide the experimental and theoretical basis for osteosarcoma treatment and prognosis. Method: 1 person osteosarcoma cell line Saos 2 cell immunofluorescence staining. Blank group served as controls. BIP protein expression was observed with a fluorescence microscope. The 2 person osteosarcoma cell line Saos 2 cell extraction of total cellular RNA by RT-PCR kit for PCR reaction. Detecting whether there The expression BipmRNA. 3 to take clinical osteosarcoma its surrounding normal tissue specimens of 20 cases, osteosarcoma without metastasis in 6 patients, 14 cases of metastasis; preoperative chemotherapy were 12 cases, eight cases of preoperative chemotherapy. Do paraffin sections for immunofluorescent staining. Detection Bip expression intensity. Results: 1 Bip in human osteosarcoma cell line Saos 2 cell expression, mainly in the endoplasmic reticulum, the cell cytoplasm was stained as green as positive. Stained dark green for the whole cytoplasm shallow class bubble-like cytoplasmic green dye. 2BipmRNA expression in human osteosarcoma cell line Saos 2 cells. 3 specimens Bip expression. Bip expression intensity of tumor tissue gt; the normal tissue tumor metastasis gt; without metastasis, preoperative chemotherapy group gt; preoperative chemotherapy group. Tumor metastasis and without metastasis, significant differences in the expression levels of preoperative chemotherapy and preoperative chemotherapy group. Conclusion: This study showed that Bip can stimulate tumor proliferation, survival, metastasis, and the appropriate response to chemotherapy medication. Bip as a flesh and blood tumor growth and metastasis and response to therapy, molecular targeted marker, can also be applied in clinical work Bip as biomarkers that determine the development and prognosis of osteosarcoma. Means of using gene down Bip expression may inhibit the growth of osteosarcoma cells to increase the sensitivity of osteosarcoma cells to chemotherapeutic drugs to control their transfer and improve the prognosis. The combination therapy to inhibit Bip expression may represent new methods for eradication of residual osteosarcoma. Therefore, the use of Bip as a molecular target or medium osteosarcoma gene therapy has broad application prospects.
