Abstract


[Objective ] study of human gastric carcinoma and adjacent normal gastric mucosa PD-1 expression and cellular localization of PD- 1 expression , PD-1 T lymphocyte function , in combination with the clinical and pathological features were analyzed retrospectively . [ Method ] detected 68 cases of gastric cancer and normal tissues of PD-1 expression using immunohistochemical streptavidin- biotin - biotin - peroxidase complex method (SABC) , combined with clinical data were retrospectively . Determination of the PD-1 expression in gastric cancer cell positioning ; using immunofluorescence and flow cytometry analysis of PD-1 expression in various types of lymphocytes , and PD-1 T lymphocyte functional changes . [Results] The results of immunohistochemistry : PD-1 expression in gastric carcinoma was 77.9% ( 53 / 68 ) , the control group adjacent normal gastric mucosa of PD-1 expression was 13.2% ( 9 / 68 ) ; independent of the combination on the clinical and pathological features of PD-1 expression and depth of invasion , TNM stage , lymph node and distant metastasis (p lt; 0.05), but not with gender age , tumor size, location , and typing (p gt ; 0.05 ) ; immunofluorescence and flow cytometry results show : gastric infiltrating lymphocytes of PD-1 upregulation , and PD-1 CD8 T-cell high expression ; high expression of PD-1 CD8 T- cell capacity of its ability to add value and secretion effectors Perforin and Granzyme B is lower than normal CD8 T-cell . [Conclusion] PD-1 expression in gastric cancer was significantly higher than that in normal gastric mucosa . Gastric expression of PD-1 is mainly CD8 T cells, high expression of PD-1 in T lymphocyte proliferative capacity and secretion of cytokines capacity decreased PD-1 T lymphocyte function is suppressed .
