Abstract

For a long time, due to the lack of a simple and fast typing methods were evaluated against Candida albicans Candida albicans, which can not carry out effective epidemiological survey. Based on phenotypic typing methods are cumbersome and lack of Candida albicans sensitivity and reproducibility, the ability to identify, looking for fast, accurate typing methods identified pathogens become an urgent problem. The subject trying to understand HIV infection and healthy population of oral Candida albicans genotype distribution; whether differences exist two groups oral Candida albicans genotype; provide a theoretical basis for the pathogenesis and epidemiological studies of Candida albicans and research basis. 2 Methods In this study, the experimental group of 54 isolates Dali, Yunnan HIV-infected people oral white Candida control group of 54 separate healthy people of the same area of ??oral Candida albicans. Extracted two populations oral Candida albicans genomic DNA, and then using the CA-INT primer PCR amplification of Candida albicans contains 1 intron encoding 25S rRNA regions of the rRNA (rDNA), according to the size of the amplified bands gene typing, and apply the X2 test for two groups of oral Candida albicans genotype analysis. 3 Results 3.1 54 HIV-infected oral Candida albicans, select the CA-INT-L and CA-INT-R two specific primers, was amplified by PCR, all white Candida can be amplified by a clear and stable strip detection success rate of 100%, the smallest strip of about 450bp, the largest strip about 840bp, 45 strain has approximately 450 bp 5 strain has approximately 840bp strain also has about 450bp and 840 bp bands, which has a Type A (only 450bp), Type B (only 840bp), C (both 450bp, another 840 bp) three genotypes, type A is the most common, while the B-type and C-type rare. 3.2 in the control group of 54 healthy people strains oral Candida albicans, can also be amplified clarity and stability of the strip, wherein the minimum strip of approximately 450bp, maximum strip approximately 840bp, 36 strains having approximately 450 bp, 12 strains having about 840bp 6 at the same time, 450bp and 840 bp bands, be classified as Type A (only 450bp), B (only 840bp), C (both 450bp, 840bp of) the three genotypes, A The most common type, followed by B, C-type rare. 3.3 HIV infected people oral Candida albicans type A share of the constituent ratio (83.33%) is greater than the healthy population of oral Candida albicans type A share form (66.67%), X2 test, P lt; 0.05, difference statistics significance; HIV-infected people oral Candida albicans in the B share constitutes less than the healthy population (9.26%) oral Candida albicans type B share of the constituent ratio (22.22%), X2 test, P gt; 0.05, the difference not statistically significant; of HIV-infected people oral Candida albicans C share (7.41%) less than the healthy population oral Candida albicans a C share (11.11%), the X2 test, P gt; 0.05 The difference was not statistically significant. 3.4 HIV infected people and healthy people oral Candida albicans genotype distribution SPSS17.0X2 test results showed that: X2 = 4.282, P = 0.118 by the alpha = 0.05 level, between the two groups A, B, C, and c 'gene type distribution was no significant difference. 4 Conclusions 4.1 HIV-infected people and healthy people oral Candida albicans genotype distribution constitute the same, are A, B and C-type, so the two groups of oral Candida albicans gene polymorphism. 4.2 HIV infection by oral Candida albicans and Candida health crowd oral white genotype in the largest proportion of A-type and B-type, followed by C-less. HIV-infected people oral Candida albicans type A share of the constituent ratio greater than the healthy population oral Candida albicans type A share of the constituent ratio of the difference was statistically significant; B and Type C in the two groups oral Candida albicans Share of form than the difference was not statistically significant. 4.3 using CA-INT primers PCR method is one of the Candida albicans genotyping method, this method is more scientific and rational, simple and fast, the result is a stable, accurate and reliable, so specific CA-INT primers PCR method is a better white strains Candida species identification method is suitable for clinical and research use.
