Filter bacteria isolated from the soil , produce extracellular melanin strains screened 27 yield melanin strains 4 , and 88 strains , fermentation conditions , to determine the yield of melanin media and culture conditions . Preliminary identification of the structure of the No. 88 strains of melanin production , determine melanin by 11 kinds of ingredients , and speculated that the eight components of Formula . 321 strains isolated from soil were screened , four strains of high-yield extracellular melanin . By comparing the ability of melanin production in the different media , as a high-yield black strains determine the No. 88 bacteria . Initially identified by morphological and biochemical identification of the strains of the genus Streptomyces . In view of the high yield bacteria black package prime , can be used as a new class of melanin production strains resources . Designed a series of experiments to study the influence of the pH value, sodium ion, magnesium ion, ammonia, peptone , glucose , and tyrosine on melanin yield , determined its melanin highest yield of the medium , and their fermentation conditions optimized to determine the optimum fermentation conditions . Separated from the No. 88 strain fermentation broth extracted melanin, and purified by extraction, thin layer chromatography , silica gel column chromatography , high performance liquid chromatography, etc. methods carried out on melanin purified melanin ESI-MS detecting, determining, which consists primarily of 11 kinds of components , and the 11 kinds of components of the high resolution mass spectrometric detection , combined with existing literature and analyzed the resulting melanin structure deduced wherein eight components of the formula .
