Abstract


Background and purpose of ovarian epithelial cancer is one of the common three malignant tumors of the female reproductive system, often non-specific clinical symptoms early, patient visits have been mostly in the late mortality rate ranks first in gynecological malignancies, and 5-year survival rate hovered at 30% to 40%. Ubiquitin (Ubiquitin, Ub) and its dependent non-lysosomal pathway in tumorigenesis and development process plays an important role, it is highly conserved peptide present in all eukaryotic tissues, free molecules or ubiquitin conjugating the form of the protein (or protein ubiquitination). Ubiquitin is widely distributed in various tissues, a component is involved in abnormal protein translation modification, its main role is involved in protein degradation. Function ubiquitin ligase system - specific ATP-dependent enzyme catalyzed cell protein ubiquitination, and thus the participation of the two major lysosomal and non-lysosomal system ubiquitination cell protein was degraded. Ubiquitin - proteasome pathway that barriers will affect cell cycle regulation, and to strengthen the role of some oncogenes can make the reduced role of the tumor suppressor gene, thereby promoting tumorigenesis. Thus, the cell protein ubiquitin for many cell activity process is critical, including the selective degradation of intracellular proteins and cell stress response of a variety of noxious stimuli and cell injury protection reaction. However, Ub exists in ovarian cancer, abnormal expression of the expression changes can influence the incidence of ovarian cancer development and prognosis is not very clear. Cyclin B1 (CyclinB1) is a classic division cycle protein involved in the regulation of cell cycle checkpoint, and to promote G 2 / M phase transition and accelerated cell cycle progression. Materials and Methods 1 collected in our hospital from January 2004 to December 2007 the gynecological inpatient surgical resection of ovarian epithelial tumor paraffin-embedded tissue specimens of 70 patients and normal ovarian tissue specimens of 20 cases were not all specimens preoperative radiotherapy, chemotherapy and other treatments. III, Ⅳ of ovarian cancer tissue the Ub protein expression was significantly higher than Ⅰ, Ⅱ stage, the difference was statistically significant (x 2 = 4.529, P <0.05); G in < / sub>-stage ovarian cancer tissue Ub protein expression was significantly the above G 1 , G 2 grade the significant difference (x 2 = 3.964, P <0.05); metastatic lymph nodes in Ub protein expression was significantly higher than those without lymph node metastasis organization, the difference was statistically significant (x 2 = 6.269, P <0.05) . 3 in epithelial ovarian cancer tissues of CyclinB1 expression with histological grade, clinical stage, lymph node metastasis-associated (P <0.05); histological type was no significant correlation (P> 0.05). III, Ⅳ of ovarian cancer tissue CyclinB1 protein expression was significantly higher than Ⅰ, Ⅱ stage, the difference was statistically significant (x 2 = 5.520, P <0.05); of G 3 < / sub>-stage ovarian cancer tissue CyclinB1 protein expression was significantly the above G 1 G 2 level, the difference was statistically significant (x 2 = 4.217, P <0.05); metastatic lymph nodes the CyclinB1 protein expression was significantly higher than those without lymph node metastasis tissue, and the difference was statistically significant (x 2 = 6.226, P <0.05) . 4 ovarian cancer tissue Ub and CyclinB1 protein expression: Ub of protein and CyclinB1 protein 7 cases were positive expression in 50 cases of ovarian cancer tissue, 19 cases were negative, Ub protein expression CyclinB1 protein negative expression by 10 cases, Ub protein negative expression CyclinB1 positive expression of 14 cases, the results indicate that the expression levels of both show a high degree of consistency by the Spearman rank correlation analysis, the correlation coefficient r = 0.301, P = 0.033, indicating that Ub and CyclinB1 protein expression in ovarian cancer tissue is positively correlated. Conclusion 1. Epithelial ovarian cancer tissue from normal ovarian tissue, benign epithelial ovarian tumor tissue to of Ub and CyclinB1 protein positive expression rate gradually increased, and more expressed in later clinical stage, histological grade poor lymph node metastasis in ovarian cancer, suggesting that both high expression may contribute to the occurrence of ovarian cancer development, invasion and metastasis. In ovarian cancer tissue increased with the CyclinB1 protein increased Ub protein expression, and a positive correlation between expression of of Ub and CyclinB1 in epithelial ovarian cancer tissue, suggesting that the two may be in the process of the development of ovarian cancer There is a synergistic effect. 3.
