Abstract


Objective To study the rat liver warm ischemia reperfusion injury of the different timing of the portal vein and the hepatic artery after exploring whether thereby reducing ischemia- reperfusion injury and its possible mechanism . Methods in healthy male SD rats 96 , divided into six groups according to the random number table , a group of sham group , the remaining five groups for the experimental group : sham group ( 16 ) , only for laparotomy and hilar anatomy ; 5 experimental groups ( groups of 16 ) are grouped according to the different open timing of the portal vein and hepatic artery reperfusion : first open the portal vein 1 min with subsequent hepatic artery group , first open the portal vein 2 min and then open the hepatic artery group , first open hepatic artery 1 min , and then open the portal vein group , first open the hepatic artery 2 min and then open the portal vein group , at the same time open the portal vein and the hepatic artery . Each group at 2h after 4 h of serum ALT and AST values ??; liver tissue malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peptide (GSH) content ; pathological HE staining of liver tissue damage ; situ end labeling (TUNE Act ) detected a group of liver tissue apoptosis . The results of the sham-operated rats liver basically normal , all the indexes in each experimental group , the differences were statistically significant ( P lt; 0.01 ) . First open the portal vein in each experimental group 1 min with subsequent hepatic artery in rat liver ischemia -reperfusion injury lightest , ALT , AST , MDA and AI values ??were significantly lower than the other experimental groups , the difference was statistically significant (P lt; 0.05 or P lt ; 0.01 ) , and SOD and GSH values ??were higher than the other experimental groups , the difference was statistically significant (P lt; 0.05 or P lt ; 0.01 ) . HE staining , first open the portal vein 1 min with subsequent hepatic artery in rat liver tissue damage light compared to the other experimental groups . After the conclusion of warm hepatic ischemia by briefly open the portal vein , hepatic artery re-opening measures can reduce the rat hepatic ischemia-reperfusion injury , may reduce the generation of reactive oxygen species and protect the liver antioxidant system .
