Abstract


Objective:The renal pelvic carcinoma is a relatively uncommon urothelial malignancy, the prognosis is not good. The disease incidence of renal pelvic carcinoma has a trend of increasing. The most cases that caused the death are its invasion and metastasis. MACC1is a new gene which associated with metastasis in colon cancer. Expression of MACC1was found significantly upregulated in many malignant tissues compared to normal tissues. MACC1is a major regulator of its transcriptional target gene c-Met. c-Met activates the HGF/c-Met signaling pathway, resulting in tumorigenesis and metastasis in many cancers. Today, prognosis of patient’s survival and treatment planning is mainly based on histopathological staging. This report is to investigate the expression of MACC1and c-Met in renal pelvic carcinoma and normal mucosa by immunehistochemistry, analyze the clinical pathological features, then demonstrate the relationship with the occurrence, development, invasion and metastasis of renal pelvic carcinoma, to find the early defective markers of invasion and metastasis of renal pelvic carcinoma and to research the useful ways to depress them.Methods:The expression of MACC1and c-Met in20cases of normal mucosa and73cases of renal pelvic carcinoma was detected by immunohistochemistry, and the clinical pathological features were analyzed. All datas were analyzed by SPSS19.0statistical software package, we used the Wilcoxon test for the comparsion of the strength of the expressions, the chi-square test for the comparsion of high expression rate, the Spearmen rank correlation analysis for the strength of expressions.Results:1. The expression strength and high expression rate of MACC1in renal pelvic carcinoma tissue were significantly higher than that in normal mucosa of renal pelvis with statistically difference, respectively(Z=-3.433, P=0.001;χ2=15.161, P=0.000). Positive expression of MACC1was increased in renal pelvic carcinoma. MACC1expression strength and pathological stage, smoking were positively related(Z=-2.560, P=0.010; Z=-2.968, P=0.003), but the expression strength and sex, age, pathological degree and urine cytology showed no statistically difference. The high expression rate of MACC1and pathological stage were positively related(χ2=3.922, P=0.048), but no related with the others.2. The expression strength and high expression rate of c-Met in renal pelvic carcinoma tissue were significantly higher than that in normal mucosa with statistically difference(Z=-2.537, χ2=0.011;χ2=5.580, P=0.018). Positive expression of c-Met was increased in renal pelvic carcinoma. c-Met expression strength and pathological stage were positively related(Z=-2.958, P=0.003), but the expression strength and sex, age, pathological degree, smoking and urine cytology showed no statistically difference. The high expression rate of c-Met and pathological stage were positively related(χ2=5.074, P=0.024), but no related with the others.3. There was a positive correlation between the expression strength of MACC1and c-Met in renal pelvic carcinoma(r=0.573, P=0.000).Conclusions:1. The expression strength and high expression rate of MACC1and c-Met in renal pelvic carcinoma tissue were significantly higher than that in normal mucosa with statistically difference, the over-expression of MACC1and c-Met were relevant with the development and progression of renal pelvic carcinoma.2. The expression strength and high expression rate of MACC1and c-Met were positively related with pathological stage, they may play an important role in invasive and metastasis of renal pelvic carcinoma.3. The expression strength of MACC1were positively related with smoking, but the high expression rate of MACC1were no related with smoking, the relationship between smoking and MACC1, renal pelvic carcinoma need to be further studying.4. Positive correlation was observed in renal pelvic carcinoma between MACCl and c-Met, it suggested that MACC1and c-Met may play a synergetic effect in the development, progression and metastasis of renal pelvic carcinoma.5. The combined detection of MACC1and c-Met can be used to determine the biological behavior and prognosis of renal pelvic carcinoma.
