Studies on the Expression of Bmi-1 and C-myc in Non-small Cell Lung Cancer and Their Correlation

Abstract


Objective To investigate the expression of Bmi-1, C-myc protein and Bmi-1 gene in non-small lung cancer tissues and away-from-cancer normal tissues, to explore the relation to clinic pathological characters and their correlation, and to prove some new basis on ocurrance, development and prognosis of NSCLC.Methods①The expression of Bmi-1 and C-myc protein was detected immunohistochemistrically in 52 cases of NSCLC and 30 cases of corresponding away-from-cancer normal tissue.②The expression of Bmi-1 mRNA was detected by RT-PCR method in 30 cases of NSCLC and 20 cases of corresponding away-from-cancer normal tissue.Results①Results of Immunohistochemistry:The expression of Bmi-1 was found mainly in nucleus, few in cytoplasm, and C-myc only in nucleus. The expression of Bmi-1 and C-myc protein in NSCLC was significantly higher than that in away-from-cancer normal tissue (Z=-4.837、-5.907, P<0.01); The positive expression of Bmi-1 protein was closely correlated with TNM staging and lymph node metastasis (Z=-2.567、-2.366, P<0.05); and C-myc protein expression was correlated with differentiation, TNM staging and lymph node metastasis (Z=-2.193、-2.596、-2.056, p<0.05); Both of them were not related to gender, age of patient, type of carcinoma (P>0.05).②Correlation analysis:Positive correlation was found between the expression of Bmi-1 and C-myc protein in NSCLC (r=0.463, P<0.01).③the result of RT-PCR:The expressive quantities of Bmi-1 mRNA in NSCLC was obviously higher than that in away-from-cancer normal tissue (t=5.188, P<0.01). The expressive quantity of Bmi-1 mRNA was closely correlated with TNM staging and lymph node metastasis:the expression of it in stageⅢ~Ⅳof NSCLC was higher than that in stageⅠ~Ⅱ(t=-2.797, P<0.05); and Bmi-1 mRNA in NSCLC with lymph node metastasis was obviously higher than that without lymphnode metastasis (t=5.617, P<0.01). But no difference was found in age, sex, differentiation, tumor size and histotype group (P>0.05).Conclusions:①The expression of Bmi-1 and C-myc may indicate that they are important factors in NSCLC ocurance.②Both Bmi-1 and C-myc express strong either in stage III-IV or with lymph node metastasis may reveal that they are also important in tumor infiltration, progression, and metastasis.③Positive correlation is found between Bmi-1 and C-myc protein in NSCLC may suggest that over-expression of Bmi-1 and C-myc protein has synergetic effect in the process of occurrence and development of NSCLC.④Whether Bmi-1 and C-myc can serve as target spots in the molecular therapy of NSCLC need further research.
