Genotypic Analysis of Hepatitis C Virus Isolated from Yantai of Shandong

Abstract


Objective To understand the hepatitis C virus (HCV) genotype in Yantai district of Shandong province, and explore whether the HCV genotypes was relevant to the liver injury through the index of liver function.Methods Specific PCR primers were taken to amplify the HCV RNA 5’UTR and/or NS5B, then PCR products were sequenced. With the GenBank reference sequence alignment, samples were genotyped combined with the phylogenetic tree.Results There were 9 cases could be genotyped in 22 HCV-RNA-positive blood donation samples, the rate was 40.9%; there were 33 cases could be genotyped in 48 HCV-RNA-positive patients, the rate was 68.8%. Among 9 blood donors we detected two kinds of genotypes,1b (8 cases,8/9,88.9%) and 3a (1case,1/9,11.1%). Among 33 cases of hepatitis C patients we detected three kinds of genotypes, lb (22 cases, 22/33,66.7%),2a (10 cases,10/33,30.3%) and 6a (lease,1/33,3.03%). Subtype 1b is the advantage genotype in HCV carriers from Yantai district, and the distribution of 1b was no significant difference in the different population (χ2=0.796, P=0.373); The difference of indicator of liver damage in the different genotypes of subjects were significant (P<0.05), the mean value of ALT, AST of 2a-subtype carriers was significantly higher than that of lb-subtype population.Conclusion The genetic diversity of HCV in Shandong Yantai is presented. The main genotype is lb-subtype, and 3a and 6a-subtype are detected firstly; There are latent HCV latent infection in population, HCV detection in donors should be enhanced to ensure the clinical safety of blood transfusion; The genotypes of HCV are relevant to the liver damage indicators,2a-subtype hepatitis C virus infection may play an important role in the poor development of liver cells.
