Abstract


Dendrin belongs to a new class of dendritic proteins, and its expression can be regulated by prolonged behavioral activity. Previous studies have been focused on brain and kidney, however whether Dendrin exists in primary sensory neurons is still unknown. In present study, using immunohitochemistry we identified the locolization and regulation of Dendrin in dorsal root ganglia (DRGs) and spinal dorsal horn under normal condition and after peripheral nerve injury. Fourteen days after sciatic nerve transection (axotomy), Dendrin-like immunoreactive fibers were increased in ipsilateral dorsal horn, mainly in laminaⅠtoⅢ, as compared to contralateral dorsal horn. These positive fibers may originate from middle and large-sized DRG neurons. We also found that Dendrin expressed in the cell bodies and the axons of DRG neurons, mainly in small-sized neurons. The number of Dendrin-positive neurons was upregulated in ipsilateral DRGs, whereas the intensity of Dendrin like-immunoreactivity was reduced. Moreover, more middle and large-sized DRG neurons were Dendrin positive in the ipsilateral DRGs as compared to the contralateral ones.The regulation of Dendrin in DRGs and spinal dorsal horn after nerve injury indicates that Dendrin may play a role in the sensory system. Nerve injury can induce neuropathic pain which is modulated by complex molecular pathways. Therefore, studying expression and regulation of Dendrin, specially identifying its role in conducting sensory singals, may provide more information to understand neuropathic pain.
