Abstract


ObjectiveTo investigate by immunochemistry (IHC) the expression level and significance of Msx2 and topoⅡ-a in sinonasal inverted papilloma(SNIP),and the relationship in the process of SNIP malignant transfomation.MethodsImmunohistochemical method was used to detect the expression of Msx2 and topoⅡ-a in 32cases of sinonasal inverted papilloma (SNIP),30 cases of sinonasal squamous cell carcinoma(NSCC) and 30 cases of inflammatory nasal polyp(INP) paraffin tissue.According to the pathology results SNIP were divided into mild dysplasia(10cases), moderate dysplasia(14cases), severe dysplasia(8cases).And Positive values of cells were measured by manual counting method and image analysis (IA) technology. All the datas were analysised by spss13.0, P<0.05 was refered to statistically significant difference.ResultsThe positive expression of Msx2 and topoⅡ-a in the NSCC were all 100%(30/30), were significantly higher than the SNIP and INP, and the pairwise comparisons were all statistically significant (p<0.05).The mean optical density (±standard deviation) of Msx2 in SNIP, NSCC tissues were 0.2183±0.5975 and 0.2521±0.0761, were significantly higher than the INP tissue 0.1878±0.03716; (P<0.05). The mean optical density (±standard deviation) of topoⅡ-a in SNIP, NSCC tissues was 0.2303±0.0397,0.2666±0.0483, were significantly higher than the SINP tissue 0.1978±0.0388 (P<0.05). The expression of Msx2 and topoⅡ-a in the SNIP which was divided according to pathological, using of manual counting method analysis:only mild dysplasia and severe dysplasia group had significant difference (P<0.05), while the light degree of dysplasia and moderate dysplasia, moderate dysplasia and severe dysplasia were all not statistically different between; using of IA systems analysis: three pathologic analysis of differences between any two groups were statistically significant. And the expression of Msx2 and topoⅡ-a was positively correlated in SNIP (P<0.05).ConclusionsMsx2 and topoⅡ-a may play an important role in the pathway of progression of SNIP and NSCC, which may be identified as the new therapeutic targets. Msx2 may have a synergic role with topoⅡ-a in malignant transfomation of SNIP.
