The Expression of HSF2 and ApoC-Ⅲ in Colonic Mucosa of Patients with Ulcerative Colitis

Abstract


Background:Ulcerative colitis is one of the inflammation bowel disease, which is a chronic nonspecific inflammation of the colon disease. Although the true pathogenesis of it is still unknown, there are evidences that many ingredients including environment, inheritance, infection, allergy, psychological factors, and some immunological factors may be involved in its development. It is widely believed that immunological mechanisms play important roles in the process. Others of our research group have used the two-dimensional gel electrophoresis(2-DE) technical to analysis the serum proteome of ulcerative colitis patients and healthy volunteers, and the technology of mass spectrometry and the methods of bioinformatics were combined to identify the differentially expressed proteins in Ulcerative colitis. They screened and identificated nine serum specific differences proteins of UC successfully, in which HSF2 and Apo C-Ⅲare expressed upward.Objectives:We aimed to investigate the expression of heat shock factor 2 (HSF2) and Apolipoprotein C-Ⅲ(ApoC-Ⅲ) in colonic mucosa in UC and study the relationship between the three parameters and disease activity.Methods:Thirty eight patients with clinically and endoscopically determined UC were studied, which was divided into mild group(13 cases)and moderate group(10 cases) or severe group(15 cases).We chose ten normal colonic mucosa as controls. The expression and localization of HSF2 and ApoC-Ⅲwere examined in colonic biopsy specimens by immunohistochemistry. Positive cells of the sections were counted by Image Analysis System. Differences among groups were compared by SPSS17.0 statistics software using t test. P<0.05 was selected as significant standard.Results:①The expression of HSF2 in UC’bowel mucosal tissue was higher than the normal group(44.4±10.7 vs 30.2±6.3), P<0.01.The express strength of HSF2 was connected with the severity of illness, and There were significant differences between mild group, moderate group and severe group (47.3±7 vs 58.5±10.8 vs 76.5±8.7).The severe group’expression of HSF2 was higher than the mild group and moderate group (p<0.05),while the moderate group’was higher than the mild group (p<0.05).②The expression of Apo C-Ⅲin UC’bowel mucosal tissue was higher than the normal group (44.4±10.7 vs 30.2±6.3), P<0.01. The express strength of ApoC-Ⅲwas connected with the severity of illness too, and there were significant differences between mild group, moderate group and severe group (53.5±11.2 vs 43.1±6 vs 37.5±6.7), The mild group’expression of ApoC-Ⅲwas higher than the moderate group and severe group (p<0.05),while there were no different between the moderate group and the severe group (p=0.109)Conclusions:①The UC group’expression of HSF2 was higher than the control group, and it’s refer to the severity of illness. The severer was UC, the more was expression. It point out that HSF2 may be connected with UC,and participated in the development of UC. The expression reflect inflammation degree of colonic mucosal,and it’s expression level can be made to judge the severity of UC and the norm of curative effect.②The expression of ApoC-Ⅲin UC’bowel mucosal tissue was higher than the normal group. It mainly expressed in inflammatory cells and glandular epithelium cells. The express strength of ApoC-Ⅲwas connected with the severity of illness, and the mild group’expression of ApoC-Ⅲwas higher than the moderate group and severe group. It point out that ApoC-Ⅲmay be connected with the development of UC. ApoC-Ⅲmaybe appear a function with the Local inflammation aggravating.③The specific mechanism of HSF2 and ApoC-Ⅲisn’t clear currently, and it needs to research further more about the activation and adjusting process of HSF2 and ApoC-Ⅲ.
