Abstract

Objective: To establish different types of intestinal bowel syndrome (IBS) animal models, using immunohistochemical methods to detect the intestinal mucosa of PAR-2 ??expression, understand the differences in the expression of PAR-2 ??in the the different IBS rat model of intestinal mucosa further explore the possible pathogenesis of IBS increased visceral sensitivity. Methods: 30 adult male SD rats were randomly divided into constipation-predominant irritable bowel syndrome (C-IBS), diarrhea-predominant irritable bowel syndrome (D-IBS) and normal control group of group 3, n = 10 using ice the the saline gavage manufacturing animal model of C-IBS, D-IBS animal models and restraint stress manufacture acetic acid enema. Distension reflex test (CRD) in each group are used to assess its visceral sensitivity and detection rats stool and gastrointestinal transit time, to evaluate the histological examination of the histological changes of the intestinal mucosa. Then detected using immunohistochemistry PAR-2 ??expression in the intestinal mucosa of different subtypes of IBS rat model group and control group, and statistical analysis to understand PAR-2 ??expression and IBS subgroups relationship. Results: (1) in the rectal distension, D-IBS group rectum minimum capacity expansion and contraction threshold (TVAC) was significantly less than the control group and the C-IBS group (P <0.05). 0.8,1.2,1.6 mL capacity expansion, the number of D-IBS group abdominal contraction significantly more than the control group (?) (?) C-IBS group (P <0.05). C-IBS group rectal expansion and contraction the smallest capacity threshold is higher than the D-IBS group and the control group (P <0.05), and significantly reduced the number of C-IBS group rat abdominal wall contraction expansion of low-volume (0.8 mL), and the control group and D-IBS group there is a statistically significant difference (P <0.05, Table 2). (2) The model rats stool detected: D-IBS group bound stress period (3h) rat feces were soft stools or mushy stools, a significant number of fecal pellets and fecal wet blank control group (P <0.01 ), fecal water content was significantly more than the control (P lt; 0.05), D-IBS group was significantly shorter in the blank control group (?) (?) will be discharged by the ink-stained black C-IBS group (P <0.05) . C-IBS group 12h number of fecal pellets and fecal wet weight were significantly less than the control group (P <0.01), fecal water content and blank control group decreased (P <0.05), C-IBS group ink-stained black. The discharge time is significantly longer than the control group (P <0.05, Table 1). (3) colon histological analysis showed that the rats in each group colonic mucosa without obvious pathological changes, in line with the characteristics of functional gastrointestinal disorders (1,2,3). (4) D-IBS group PAR-2 ??positive cells than intensive, its IOD value of 58.69 ± 8.422, while the C-IBS group the IOD value is 33.2 ± 4.237, control group the IOD value is 29.5 ± 3.206. D-IBS group colon tissue PAR-2-positive cell number was significantly more than the control group (?) (?) C-of IBS (P <0.05), C-of IBS group and the control group of PAR-2 ??cell number no noticeable significant difference (P > 0.05, as shown in Figure 4, 5, 6, Table 3) Conclusion: (1) acetic acid enema plus restraint stress and ice saline gavage method made IBS animal model are in line with visceral hypersensitivity of IBS, and histological analysis Displays the colon tissue of each model group had no significant pathological changes. Can be used to experiment with IBS research. (2) made of two methods of animal models stool and fecal gastrointestinal time detection characteristics of diarrhea, constipation, IBS diarrhea, constipation-animal model can be used for typing. (3) in D-IBS rat colon tissue PAR-2 ??positive cells significantly more than the control group and the C-IBS, prompted a certain PAR-2 ??changes with different subtypes pathogenesis.
